Objective To investigate the expression level and significance of long non-coding RNA(lnc-DC)in dendritic cells(DC)in primary immune thrombocytopenia(ITP).Methods A total of 88 adult patients with primary ITP and 60 healthy subjects(control group)were collected in the First People's Hospital of Xianyang,and the patients with ITP were divided into effective group and refractory group.Real-time quantitative PCR(qRT-PCR)was used to detect the relative expression of Lnc-DC in the blood samples of each group.The surface CD80 and CD86 of peripheral blood mononuclear cells were measured by flow cytometry.Interleukin-6(IL-6),IL-23 and Th1 and Th2 cytokines IL-2 and IL-4 in the supernatant of activated DC cells in each group were determined by enzyme-linked immunosorbent assay.The x2 test,one-way ANOVA,Mann-whitney test and Spearson method were used to analyze the data.Results ① The platelet count(PLT)and IL-4 level of the effective group and the refractory group were significantly lower than the control group,and those in the refractory group were lower than the effective group[PLT:(95±24)×109/L vs.(31±9)×109/L vs.(189±48)× 109/L;F=280.82,P<0.001;IL-4:(3.2±0.6)pg/ml vs.(1.3±0.4)pg/ml vs.(4.5±0.6)pg/ml;F=375.76,P<0.001].②The levels of lnc-DC、CD80、CD86、IL-6、IL-23、IL-2 in the effective group and the refractory group were significantly higher than those of the control group,and those in the refractory group were higher than the effective group lnc-DC:[(2.7±0.5)pg/ml vs.(5.6±0.5)pg/ml vs.(2.4±0.4);F=702.60,P<0.001;CD80:(40.3±4.1)%vs.(62.1±4.2)%vs.(36.9±2.6)%,F=316.75,P<0.001;CD86;[(46.6±5.5)%vs.(65.6±5.4%)vs.(40.7±4.9)%;F=316.70,P<0.001;IL-6:(83±24)pg/ml vs.(165±27)pg/ml vs.(60±10)pg/ml;F=321.25,P<0.00l],IL-23[(29±8)pg/ml vs.(83±16)pg/ml vs.(14±3)pg/ml;F=605.14,P<0.00l;IL-2:[(5.7±1.8)pg/ml vs.(13.0±2.4)pg/ml vs.(3.0±0.9)pg/ml;F=371.14,P<0.001].③ The relative expression level of Lnc-DC in serum of patients with ITP was negatively correlated with platelet count and IL-4 level(r=-0.52,-0.77,both P<0.05).The relative expression level of LCC-DC in serum was positively correlated with the levels of CD80,CD86,IL-6,IL-23 and IL-2(r=0.42,0.75,0.74,0.79,0.72,all P<0.05).Conclusion Serum NRC-DC is significantly up-regulated in adult patients with ITP,and its expression level is related to ITP disease,DC formation,activation and T cell immune response.
