Applications of key molecules targeting DNA damage response in tumor radiosensitization
Radiotherapy,an important method for clinically treating tumors,causes serious damage to tumor cells primarily by destroying DNA double strands.However,the outcomes of radiotherapy alone are limited by tumor cells'inherent capacity for DNA damage repair.Multiple studies have shown that key molecules targeting DNA damage response(DDR)can effectively inhibit DNA damage repair while synergistically enhancing radiotherapy and chemotherapy sensitivity.This study reviews the applications of some key DDR inhibitors combined with radiotherapy or chemotherapy in the treatment of various tumors,elaborates on cyclin guanine nucleotide adenine nucleotide synthase-interferon gene stimulator(cGAS-STING)-mediated immune responses induced by combination therapy,and finally summarizes and predicts the challenges and development prospects of combination therapy.
