Hyperbaric oxygen inhibits the malignant biological behaviors of lung cancer A549 cells by down-regulating OTX1 gene expression
Objective To investigate the effects of hyperbaric oxygen(HBO)assisted vinorelbine(NVB)treatment on the malignant biological behaviors of lung cancer A549 cells and its mechanism of action.Methods After the completion of human non-small cell lung cancer A549 cell culture,the cells were divided into NVB group and HBO+NVB group,according to the experimental design.The NVB group was treated with 50 µmol/L(IC50 concentration)of NVB,while the HBO+NVB group was treated with 50 μmol/L NVB and then 0.2 MPa HBO.CCK-8 was performed at 24,48,and 72 hours to detect the proliferation ability of lung cancer A549 cells in both groups.After 72 hours of treatment,Transwell chamber assay was used to detect cell migration and invasion ability,and Western blotting assay was used to detect the expression levels of proliferation,migration,and invasion related proteins in A549 cells.Results The expression level of OTX1 mRNA in lung cancer A549 cells in the HBO+NVB group was significantly lower than that in the NVB group(P<0.05).The proliferation,migration,and invasion abilities of lung cancer A549 cells in the HBO+NVB group were significantly lower than those in the NVB group(P<0.05).The expression levels of proliferation related proteins,i.e.,MMPs and VEGF,in lung cancer A549 cells in the HBO+NVB group were significantly lower than those in the NVB group(P<0.05).The expression level of E-cadherin,a migration and invasion related protein,was significantly increased in the HBO+NVB group,while the expression level of N-cadherin was significantly decreased(P<0.05).Conclusion Hyperbaric oxygen can inhibit the proliferation,migration,and invasion ability of lung cancer A549 cells by downregulating OTX1 gene expression.
