Objective The purpose of this study was to elucidate the expression difference of Galectin-9 in peripheral blood and colorectal cancer tissues and normal colorectal tissues and its correlation with immune cell infiltration,and to explore its effect on the proliferation,migration and invasion of colorectal cancer.Methods The expression levels of Galectin-9 in colorectal cancer tissues and normal tissues were analyzed by using TCGA database and UALCAN website.TIMER database was used to analyze the relationship between Galectin-9 and immune cell infiltration in colorectal cancer tissues.The expression of Galectin-9 in peripheral blood and tissues was analyzed by ELISA and immunohistochemistry.In two human colon cancer cell lines,HCT116 and SW480,we overexpressed Galectin-9 with recombinant plasmid,and then analyzed Galectin-9 protein levels by Western blot.Finally,the effects of Galectin-9 on the proliferation,migration and invasion of colorectal cancer cells were investigated by CCK-8 assay,scratch assay and Transwell assay.Results Galectin-9 was low in peripheral blood,tumor tissue and colorectal cancer cell line.Galectin-9 is positively correlated with immune cell infiltration of CD4+T cells,CD8+T cells and B cells in colorectal cancer.Galectin-9 can inhibit the proliferation,migration and invasion of colorectal cancer cells.Conclusion Galectin-9 is low expression in colorectal cancer,and inhibits the proliferation,migration and invasion of colorectal cancer cells,which may be related to the immune mechanism of tumors.
