摘要
目的 评价右美托咪定对大鼠肠缺血再灌注损伤时聚腺苷二磷酸核糖聚合酶-1(PARP-1)表达的影响.方法 清洁级健康成年雄性SD大鼠18只,体质量200~240 g,8~12周龄,采用随机数字表法分为3组(n=6):假手术组(S组)、肠缺血再灌注组(I/R组)和肠缺血再灌注+右美托咪定组(I/R+Dex组).S组仅开腹分离肠系膜上动脉,不夹闭;I/R组和I/R+Dex组采用夹闭肠系膜上动脉45 min恢复再灌注120 min的方法制备大鼠肠缺血再灌注损伤模型;3组均分离右侧股静脉并置管,I/R+Dex组于夹闭前静脉输注右美托咪定5 μg/kg,持续15 min,S组和I/R组给予等容量生理盐水.于再灌注120 min时,麻醉后处死大鼠取小肠组织,确定湿质量/干质量(W/D)比值;采用HE染色法观察小肠组织病理学结果并进行Chiu评分;采用TUNEL法观察肠上皮细胞凋亡情况,计算凋亡指数;采用Western blot法检测小肠组织PARP-1活化产物聚腺苷二磷酸核糖(PAR)和caspase-3的表达.结果 与S组比较,I/R组和I/R+Dex组W/D比值、Chiu评分和凋亡指数升高,PAR和caspase-3表达上调(P<0.05);与I/R组比较,I/R+Dex组W/D比值、Chiu评分和凋亡指数降低,PAR和caspase-3表达下调(P<0.05).结论 右美托咪定减轻大鼠肠缺血再灌注损伤的机制可能与抑制PARP-1活化,减轻细胞凋亡有关.
Abstract
Objective To evaluate the effect of dexmedetomidine on the expression of polyade-nosine diphosphate ribose polymerase-1(PARP-1)during intestinal ischemia-reperfusion(I/R)injury in rats.Methods Eighteen clean-grade healthy male Sprague-Dawley rats,weighing 200-240 g,aged 8-12 weeks,were divided into 3 groups(n=6 each)using a random number table method:sham operation group(S group),intestinal I/R group(I/R group)and intestinal I/R+dexmedetomidine group(I/R+Dex group).In group S,only the superior mesenteric artery was isolated by laparotomy without clipping.The model of in-testinal I/R injury was established by clamping the superior mesenteric artery for 45 min followed by reperfu-sion for 120 min in anesthetized animals in I/R group and I/R+Dex group.The right femoral vein was isola-ted and catheterized in all the three groups.In I/R+DEX group,dcxmedetomidine 5 μg/kg was intravenously infused for 15 min before clamping.The equal volume of normal saline was given in S group and I/R group.At 120 min of reperfusion,the rats were sacrificed after anesthesia and the small intestine tissues were taken for determination of the wet/dry lung weight(W/D)ratio,apoptosis in intestinal epithelial cells(using TUNEL method)and expression of PARP-1 activation product polyadenosine diphosphate ribose and caspase-3(by Western blot)and for microscopic examination of pathological changes which were assessed and scored according to Chiu.The apoptosis index was calculated.Results Compared with S group,the W/D ratio,Chiu score and apoptosis index were significantly increased,and the expression of polyadenosine diphosphate ribose and caspase-3 was up-regulated in I/R group and I/R+Dex group(P<0.05).Compared with I/R group,the W/D ratio,Chiu score and apoptosis index were significantly decreased,and the expres-sion of PAR and caspase-3 was down-regulated in I/R+Dex group(P<0.05).Conclusions The mechanism by which dexmedetomidine alleviates intestinal I/R injury may be related to inhibiting PARP-1 activation and alleviating cell apoptosis in rats.
