在线样本量计算简易工具在随机对照试验中的应用效果评价

目的 评价在线样本量计算简易工具计算结果的准确性,探讨其在临床研究环境中的优势及应用价值.方法 基于2019年9月1日-2020年12月1日自主研发的汉化在线样本量计算工具,以随机对照试验为设计类型,以PASS软件计算结果为标准,基于3项真实临床研究项目的样本量计算参数,每次通过调整单一参数形成不同的参数组合,最终定性资料和定量资料分别形成60个和80个参数组合纳入样本量计算,比较在线样本量计算工具计算结果与PASS软件计算结果的绝对差值,并绘制Bland-Altman曲线,再计算组内相关系数,评价两种工具计算结果的一致性.结果 对于定性资料的差异性检验和优效性检验,两种工具计算所得样本量几乎完全一致;对于定性资料的等效性检验和非劣效性检验,两种工具计算所得样本量差异在0-6之间;对于定量资料,上述四种假设检验两种工具计算所得样本量差异均在0-4之间.根据比较的Bland-Altman图结果所示,两种工具计算的样本量结果均值和差值所形成的散点几乎均落入95%一致性界限内,同时两种工具计算所得样本量的组内相关系数值均趋近于1,且P<0.001,达到统计学显著差异.结论 自主研发的在线样本量计算工具使用简便,计算结果准确可靠,适于辅助临床医护人员开展相关临床研究.
Evaluation of the Application Effect of Simple Tool for Online Sample Size Calculation in Randomized Controlled Trial
Objectives To evaluate the accuracy of the calculation results of the simple online sample size calculation tool,and explore its advantages and application value in the clinical research environment.Methods We developed a Chinese online tool for sample size calculation,and compared the results with PASS software.Randomized Controlled Trial(RCT)was used as the design type.Different parameters were tested.Consistency between the online tool and PASS was evaluated by absolute difference,bland-Altman curve,and Intraclass Correlation Coefficient(ICC).Results The study calculated the parameters based on the sample size of 3 real clinical research projects.By adjusting one parameter at a time,different parameter combinations were formed,finally,60 and 80 parameter combination data were obtained from qualitative data and quantitative data,respectively,and were included in the sample size calculation.For the equivalence test and non-inferiority test of qualitative data,the differences in sample size calculated by the two tools ranged between 0 and 6.For quantitative data,the differences in sample size calculated by the two tools always remained between 0 and 4.At the same time,the ICC values of the sample sizes calculated by the two tools were close to 1,which is statistically significant.Conclusions The online sample size calculation tool had good accuracy and stability,and was easy to use.It was suitable for assisting clinical medical staff in scientific research.
