2型糖尿病大鼠Roux-en-Y手术造模及技术改进

Establishment and improvement for rat model of type 2 diabetes mellitus treated by Roux-en-Y gastric bypass operation

目的 采用SD大鼠建立手术治疗2型糖尿病的实验动物模型.方法 将40只成年SD大鼠随机分成造模组(n=30)和正常对照组(NC,n=10),造模组大鼠经高脂高糖饲料喂养及小剂量链脲佐菌素腹腔注射建立2型糖尿病大鼠模型.将造模成功的28只大鼠随机分为3组,分别为Roux-en-Y旁路手术组(RYGB,n=10)、假手术组(SO,n=10)和非手术组(NO,n=8).结果 造模组大鼠血糖显著升高,均值大于16.7 mmol/L,胆固醇、三酰甘油均显著高于对照组,2型糖尿病大鼠造模成功;RYGB组大鼠血糖、总胆固醇及三酰甘油水平在术后1周即开始下降,术后4周血糖接近正常水平;改良手术技术,加强围术期管理后RYGB组大鼠术后总生存率达到80%,术后第8周,4组大鼠均长期存活.结论 改良后的2型糖尿病大鼠Rou-xen-Y旁路手术造模技术可行,有助于临床评估手术治疗糖尿病的远期效果和机制研究.
Objective To establish a rat model of type 2 diabetes mellitus treated by Roux-en-Y gastric bypass operation.Methods Forty adult SD rats were randomly divided into two groups:model group (n =30) and normal control group (NC,n =10).Model group rats were subjected to streptozotocin (STZ) intraperitoneal injection 4 weeks after feeding on high-fat and high-sugar diet.Rat model of type 2 diabetes mellitus was established successfully after detection of increased blood glucose,cholesterol and triglyceride in model group as compared with those in control group.Twenty-eight rats with type 2 diabetes mellitus were randomly divided into 3 groups:Roux-en-Y gastric bypass operation group (RYGB,n =10),sham operation group (SO,n =10),no operation group (NO,n =8).Results The blood glucose (> 16.7 mmol/L averagely),cholesterol and triglyceride levels were increased obviously in model group as compared with those in control group.Meanwhile,"eat more,drink more,urine more and weight loss" occurred in model group,suggesting the successful establishment of rat model of type 2 diabetes mellitus.The blood glucose,cholesterol and triglyceride levels in RYGB group were decreased at first week after operation,and nearly reduced to normal level at 4th week after operation.The overall survival rate in RYGB group was increased to 80% by improved operation technique and enhanced perioperative management.All rats in 4 groups were alived at 8th week post-operation.Condusion This improved technique is feasible for establishment of rat model of type 2 diabetes mellitus treated by Roux-en-Y bypass operation.This model can be observed and studied for long time,and will contribute to evaluation of long-term effect of operation on type 2 diabetes mellitus and give more help in mechanism research.
