摘要
目的 探讨Tip27基因在甲状腺癌与良性肿瘤组织中的表达差异及其与临床病理特征的相关性.方法 采用实时荧光定量聚合酶链反应(FQ-PCR)和酶联免疫吸附试验(ELISA)法分析96例肿瘤组织及癌旁组织中Tip27基因的mRNA和蛋白表达水平;对肿瘤样本中Tip27基因表达诊断特异性进行受试者工作特征(ROC)曲线分析;对Tip27基因表达与临床病理特征相关程度进行Logistic回归分析等.结果 41例甲状腺癌患者癌组织中24例Tip27基因mRNA表达较癌旁组织低,占58.5%.55例甲状腺良性肿瘤患者肿瘤组织中39例Tip27基因mRNA表达较肿瘤旁组织高,占70.9%,差异有统计学意义(P=0.004);14例甲状腺乳头状癌组织中8例Tip27蛋白表达较癌旁组织低,占57.1%;13例结节性甲状腺肿患者肿瘤组织中11例Tip27蛋白表达较肿瘤旁组织高,占84.6%,差异有统计学意义(P=0.046).41例不同临床分期甲状腺癌患者Tip27基因转录差异明显(P =0.008),优势比(OR)=2.690,为保护性因素.结论 Tip27基因在甲状腺癌组织中的表达下调,在良性肿瘤组织中的表达较癌旁组织增高,且与肿瘤临床分期呈负相关,提示其可能参与了甲状腺癌的发展与转移.
Abstract
Objective To investigate the expression difference and the correlation with clinical pathology classification of Tip27 gene in thyroid cancer and benign tumor tissue.Methods Tip27 gene and protein were detected in 96 cases of tumor and pericarcinous tissue by real-time fluorescent quantitative polymerase chain reaction (FQ-PCR) and enzyme-linked immunosorbent assay (ELISA).The diagnostic specificity of Tip27 gene expression in the tumor samples was evaluated by receiver operating characteristic (ROC) curve analysis.The related degree of Tip27 gene expression and the clinical stage was evaluated by Logistic regression analysis.Results There were 24 cases displayed lower Tip27 gene mRNA expression than those of tissue adjacent to carcinoma in 41 thyroid cancer cases,accounting for 58.5%.However,there were 39 cases displayed higher Tip27 gene mRNA expression than those of tissue adjacent to tumor in 55 thyroid benign tumor,accounting for 70.9%.There was significantly difference between carcinoma and benign tumor (P =0.004).There were 8 cases displayed lower Tip27 protein expression than those of tissue adjacent to carcinoma in 14 thyroid papillary cancer,and 11 cases displayed higher Tip27 protein expression in 13 thyroid nodular goiter cases.There was significantly difference between car cinoma and benign tumor (P =0.046).There was significantly Tip27 transcription expression difference in 41 thyroid cancer patients with different clinical stage (P =0.008),the odds ratio (OR) =2.690,and the Tip27 looks like a protective factor.Conclusion Tip27 gene expression is down-regulated in thyroid carcinoma tissue,but higher in benign tumor tissue than in tissue adjacent to the tumor,also displayed negatively correlated with clinical stage.This study speculate that Tip27 gene may be associated with thyroid carcinoma development and metastasis.
