The expression and clinical significance of circular RNA in peripheral blood of pancreatic ductal adenocarcinoma
Objective To investigate the expression levels of circular RNA(circRNA)in the peripheral blood of patients with pancreatic ductal adenocarcinoma(PDAC)and their clinical significance.Methods A total of 20 highly expressed circRNAs screened from the peripheral blood and paraffin speci-mens of PDAC patients were selected for gene knockout and cell scratch wound test,then circRNAs with statistically significant mobility were further screened.The tissues of 6 patients with PDAC and preoperative peripheral blood of 41 patients with PDAC were collected between December 2020 and June 2022 in the Third Affiliated Hospital of Soochow University.The expression of these circRNAs was detected by real-time reverse transcription polymerase chain reaction(Real-time PCR).We further analyzed the relationship between circRNAs and clinicopathological characteristics and their effects on PDAC specificity and sensitiv-ity for early diagnosis of PDAC.The t-test,non-parametric test,Chi-square test,receiver operating char-acteristic(ROC)curve and binary logistic regression were used to analyze the data.Results A total of 6 circRNAs with statistically significant mobility were screened out by cell wound scratch test.The circ_0075346,circ_0005654 and circ_0027491 were highly expressed in the peripheral blood of PDAC patients(P<0.05),in which circ_0075346 expression level was strongly correlated with lymph node metastasis(x2=6.190,P<0.05)and nerve invasion(x2=17.434,P<0.01)in PDAC patients,and circ_0005654 expression level was associated with nerve invasion(x2=6.740,P<0.01).The ROC curve analysis showed that all three circRNAs had diagnostic value for PDAC[area under curve(AUC)=0.631,0.700,0.723,P<0.05],and the combination of multiple circRNAs can improve diagnostic specificity and sensi-tivity to varying degrees.Conclusion The circ_0005654,circ_0075346 and circ_0027491 were highly expressed in the peripheral blood of patients with PDAC,and may be potential biomarkers for assessing the diagnosis and prognosis of PDAC.
