目的 观察三阴性乳腺癌细胞中长链非编码RNA linc00921靶向调控微小RNA(miR)-9-5p对细胞侵袭能力的影响.方法 收集2019年4月至2020年4月期间河北医科大学第四医院乳腺外科三阴性乳腺癌及癌旁标本共30例作为研究对象,利用实时荧光定量聚合酶链反应(PCR)分析linc00921与miR-9-5p表达的相关性;将MDA-MB-231细胞分为linc00921过表达组和对照组,利用细胞侵袭实验观察其对细胞侵袭能力的影响;利用实时荧光定量PCR检测linc00921对miR-9-5p表达的影响;采用生物信息学分析及双荧光素酶报告基因验证linc00921与miR-9-5p的关系;将细胞分为 miR-9-5p mimics+linc00921 组和 miR-NC+linc00921 组,观察 linc00921 影响细胞的侵袭能力是否与miR-9-5p有关.两组间比较采用t检验.结果 过表达linc00921组侵袭细胞数[(154.50±12.06)个]低于对照组细胞[(400.50±6.36)个,t=61.500,P<0.05];生物信息学分析及双荧光素酶报告基因验证结果表明,linc00921可与miR-9-5p结合,并降低MDA-MB-231细胞的荧光素酶活性(0.41±0.04比1.03±0.09,t=17.570,P<0.05);相关性分析结果表明linc00921与miR-9-5p 两者表达呈负相关(r=-0.484,P<0.05);挽救实验结果证明 linc00921+miR-9-5p mimics组侵袭细胞数明显高于 linc00921+miR-NC 组[(335.50±14.84)个 比(122.50±4.94)个,t=30.430,P<0.05].结论 长链非编码RNA linc00921可通过靶向调控miR-9-5p影响三阴性乳腺癌细胞的侵袭能力.
Impact of long non coding RNA linc00921 on cell invasion ability in triple-negative breast cancer via targeted regulation of microRNA-9-5p
Objective To observe the impact of long non-coding RNA linc00921 in triple-negative breast cancer cells on cell invasion ability through targeted regulation of microRNA(miR)-9-5p.Methods A total of 30 samples,including triple-negative breast cancer and adjacent tissues,from the Department of Breast Surgery at the Fourth Hospital of Hebei Medical University,collected between April 2019 and April 2020,were utilized as the research subjects.The correlation between the expression of linc00921 and miR-9-5p was analyzed using real-time fluorescence quantitative polymerase chain reaction(PCR).The MDA-MB-231 cells were divided into linc00921 overexpression group and control group,and the impact on cell invasion ability was observed through a cell invasion experiment.The effect oflinc00921 on the expres-sion of miR-9-5p was detected using real-time fluorescence quantitative PCR.Bioinformatics analysis and dual-luciferase reporter gene assay were employed to verify the relationship between linc00921 and miR-9-5p.The miR-9-5p mimics+linc00921 group and miR-NC+linc00921 group were set up,and the impact of linc00921 on cell invasion ability was observed to determine its relationship with miR-9-5p.T-test was per-formed to compare the two groups.Results The number of invasive cells in the linc00921 overexpression group(154.50±12.06)was significantly less than that in the control group(400.50±6.36,t=61.500,P<0.05).The bioinformatics analysis and dual luciferase reporter gene validation results indicated that linc00921 could bind to miR-9-5p and reduce the luciferase activity in MDA-MB-231 cells(0.41±0.04 vs.1.03±0.09,t=17.570,P<0.05).The correlation analysis results revealed a negative correlation between linc00921 and miR-9-5p expression levels(r=-0.484,P<0.05).The rescue experiment re-sults demonstrated that the number of invasive cells in the linc00921+miR-9-5p mimics group was signifi-cantly greater than that in the linc00921+miR-NC group[(335.50±14.84)vs.(122.50±4.94),t=30.430,P<0.05].Conclusion The long non-coding RNA linc00921 can affect the invasive ability of triple-negative breast cancer cells by targeting and regulating miR-9-5p.
