人源化免疫系统小鼠模型深入探索肝癌免疫机制研究进展

摘要
肝癌是全球癌症相关死亡的主要原因之一,其患病率逐年升高,特别是中晚期肝癌的预后非常差.免疫治疗是一种新型替代治疗方案.然而,由于肝癌的免疫微环境复杂,现有治疗方案只在少数人群中有效.因此,建立一个完善的人源化免疫系统小鼠模型对于深入研究肝癌进展中的免疫机制、开发新型免疫治疗方案并验证其有效性和安全性非常重要.本综述从肝癌特殊的免疫微环境出发,总结了目前不同人源化免疫系统小鼠模型的构建方法及其优缺点,包括基于造血干细胞和外周血单核细胞的模型.随后探讨这些模型在肝癌免疫治疗中的应用进展,例如肝癌机制的研究、免疫检查点抑制剂(ICIs)的安全性和有效性验证以及免疫细胞过继回输疗法等.最后,提出人源化免疫系统小鼠模型的局限性和未来发展方向,为进一步研究肝癌个体化免疫治疗提供参考.
Abstract
Liver cancer is one of the leading causes of cancer-related mortality worldwide,and its incidence rate is on the rise,especially with poor prognosis in mid to late stage liver cancer.Immunotherapy offers a potential so-lution to this problem,but its effectiveness is limited due to the complex immune microenvironment of the disease.As such,the development of a well-crafted humanized immune system mouse model is of paramount importance to facilitate the exploration of liver cancer progression,the development of novel immunotherapies,and the assessment of their safety and efficacy.This review starts from the special immune microenvironment of liver cancer and summarizes the construction methods and advantages and disadvantages of different humanized immune system mouse models,including those based on hematopoietic stem cells and peripheral blood monocytes.Furthermore,we discuss the application of these models in liver cancer immunotherapy,including research into the progression of the disease,the safety and effi-cacy of immune checkpoint inhibitors,and adoptive cell transfer therapy.Finally,we also propose the limitations and future development directions of humanized immune system mouse models,providing a theoretical reference for further research on individualized immunotherapy for liver cancer.
