Expression and clinical significance of activin A receptor type Ⅰ in gastric cancer
Objective To explore the expression,prognostic value and clinical significance of ac-tivin A receptor type Ⅰ(ACVR1)in gastric cancer(GC).Methods The transcriptome data and clinical parameter of GC were downloaded through the Cancer Genome Atlas(TCGA)database.The mRNA expres-sion of ACVR1 and the its correlation with clinicopathological features were analyzed by R.The prognostic factors of GC patients were explored by the gene expression profiling interactive analysis(GEPIA)database and Multivariate analysis.At the same time,the relevant pathways of ACVR1 were analyzed by R in order to explore its potential mechanism in GC.Results The mRNA expression of ACVR1 in GC was signifi-cantly higher[4.673(4.247,5.087)vs.3.699(3.332,4.426),Z=16.6,P<0.01],and its expres-sion was significantly correlated with the tumor invasion depth(x2=11.521,P<0.01).TCGA database analysis showed that the overall survival time/progression-free survival time in GC patients with high ACVR1 expression were shorter than in GC patients with low ACVR1 expression(P<0.01).Moreover,multivariate analysis showed that ACVR1 expression[hazard ratio=1.639,95%confidence interval:1.102-2.436,P<0.05]was an independent predictor of the prognosis of GC patients.The kyoto encyclo-pedia of genes and genomes(KEGG)pathway enrichment analysis showed that ACVR1-related differential genes were mainly enriched in the phosphatidylinositol 3 kinase(PI3K)-protein kinase B(Akt)signaling pathway,neuroactive ligand receptor interaction signaling pathway,focal adhesion signaling pathway,cal-cium signaling pathway,human papillomavirus infection pathway and other related pathways.Conclusion ACVR1 mRNA is highly expressed in GC and closely related to the poor prognosis of GC patients.Moreo-ver,it may play an important role in GC through the PI3K-Akt signaling pathway,neuroactive ligand recep-tor interaction signaling pathway,focal adhesion signaling pathway.
