The expression and clinical significance of cRAPGEF5 in gastric cancer tissue
Objective To analyze the differential expression of cRAPGEF5 in gastric cancer tissue and adjacent tissues,and analyze the expression levels of cRAPGEF5 in patients with different clinical and pathological characteristics and prognosis.Methods Totally,65 clinical specimens of gastric cancer ad-mitted to our hospital and corresponding adjacent tissues were selected as the research subjects.Total RNA was extracted and differentially expressed genes were screened for transcriptomics.Real time fluorescence quantitative PCR was used to analyze the expression levels of cRAPGEF5 in adjacent and gastric cancer tis-sues.The proliferation cell nuclear antigen(Ki-67)expression levels were detected by immunohistochemis-try in two groups.The correlation between the expression level of cRAPGEF5 and the clinical pathological characteristics and prognosis of patients was analyzed.The comparison of inter group measurement data was done by t-test.Pearson correlation analysis was used for correlation analysis.Results The transcriptome results showed a total of 458 differentially expressed circRNAs in gastric cancer tissue and adjacent tissues,of which 314 were upregulated and 144 were downregulated.This study selected cRAPGEF5 with the most significant expression difference as the research object.As compared with the adjacent tissues(2.12± 0.29),the expression level of cRAPGEF5 in gastric cancer tissue(1.21±0.17)significantly decreased(t=22.910,P＜0.05).The expression level of cRAPGEF5 in tumor tissue of patients with low Ki-67 ex-pression(2.08±0.22)and Ki-67 positive expression patients(1.76±0.21)significantly decreased(t=2.589,P＜0.05).As compared with the gastric cancer tissue of patients with moderate to high differentia-tion(2.29±0.32),the expression level of cRAPGEF5 in patients with low differentiation(1.93± 0.0.17)was significantly downregulated(t=4.231,P＜0.05).As compared with the gastric cancer tis-sue of patients with lymph node metastasis(1.80±0.08),the expression level of cRAPGEF5 in non-lymph node metastasis patients(2.16±0.19)significantly increased(t=7.126,P＜0.05).As compared with the gastric cancer tissue of stage Ⅲ-Ⅳ patients(1.82±0.18),the expression level of cRAPGEF5 in stage Ⅰ-Ⅱ patients(2.20±0.16)was significantly increased(t=5.947,P＜0.05).The 3-year survival time in patients with high expression of cRAPGEF5[(21.02±2.48)months]was significantly longer than that in patients with low expression[(10.56±2.65)months](Log-rank=5.984,P＜0.05).The clinical stage,lymph node metastasis,differentiation degree,and CRAPGEF5 level of patients with gastric cancer were related to their prognosis(P＜0.05).The multivariate Cox regression model showed that low expres-sion of cRAPGEF5 was a risk factor for poor prognosis in gastric cancer patients(P＜0.05).Conclusion The low expression of cRAPGEF5 in gastric cancer tissue is closely related to Ki-67 level,clinical stage,differentiation degree,and lymph node metastasis.The lower the expression of cRAPGEF5,the worse the prognosis of patients with gastric cancer.
