具有聚集诱导发光性能的高效增敏剂用于骨肉瘤的光热治疗研究

目的 探讨BITT的聚集诱导发光(AIE)性质,骨肉瘤细胞靶向性、荧光成像以及光热治疗效果.方法 采用MG63、143B细胞作为模型细胞,探讨其生物相容性,以商业细胞核染料Hoechst 33342为比较对象,探讨其细胞内定位区域及抗光漂白性,应用激光照射,探讨其对肿瘤细胞的暗毒性和光热治疗效果,激光照射和黑暗组两组间比较采用t检验.结果 随着激光功率的提高、浓度的升高,BITT溶液的温度迅速升高.浓度40 μmol/L的BITT溶液在激光照射153 s后可达45 ℃,相同条件下60 µmol/L可达49.5 ℃.随着激光功率的提高,浓度60 μmol/L的BITT溶液的温度迅速升高,1 W/cm2的660 nm激光照射300 s可达58.8℃.BITT即使在低浓度(20 µmol/L)下也能显示出优异的光热性能.激光共聚焦显示BITT主要定位在骨肉瘤细胞质中,具有优异的光稳定性.采用低能量(0.3 W/cm2)的660 nm激光照射,其光热治疗杀伤效果显示,在较低的浓度下(16 μmol/L),肿瘤细胞可被大比例杀伤(杀伤率>90%).结论 BITT在水溶液中具有良好的光热转换能力,其良好的光热转化能力、生物相容性、光稳定性能为后续光热治疗协同放疗增敏研究提供良好的增敏剂及基础技术支持,为骨肉瘤外科手术的协同治疗带来新的协同治疗方式选择.
Efficient sensitizers with aggregation-induced emission properties for photothermal therapy of osteo-sarcoma
Objective To investigate the aggregation-induced emission(AIE)property of BITT,and its osteosarcoma cell targeting,fluorescence imaging,and photothermal therapeutic effects.Methods MG63 and 143B cells were used as model cells to investigate their biocompatibility.Commercial nuclear dyes Hoechst 33342 were used as comparative objects to investigate their intracellular localization regions and photobleaching resistance.Laser irradiation was applied to investigate their photothermal therapeutic effects on tumor cells,comparison between laser irradiation and dark group using t-test.Results As the laser power and concentration increase,the temperature of the BITT solution rapidly increases.The con-centration of 40 μmol/L BITT solution can reach 45 ℃ after 153 s of laser irradiation,and 60 μmol/L can reach 49.5 ℃ under the same conditions.With the increase of laser power,the temperature of the BITT solution with a concentration of 60 µmol/L increased rapidly,and the 660 nm laser irradiation of 1 W/cm2 could reach 58.8 ℃ for 300 seconds.BITT could exhibit excellent photothermal performance even at low con-centrations(20 μmol/L).Laser confocal display showed that BITT was mainly located in the cytoplasm and had excellent photostability.Under low-energy 660 nm laser irradiation(0.3 W/cm2),the killing effect of photothcrmal therapy showed that at a lower concentration(16 μmol/L),tumor cells could be killed in a large proportion(killing rate>90%).Conclusion BITT has good photothermal conversion ability in a-queous solution,and its excellent photothcrmal conversion ability,biocompatibility,and photostability pro-vide a good sensitizer and basic technical support for subsequent research on photothermal therapy combined with radiotherapy sensitization,bringing new options for collaborative treatment of osteosarcoma surgery.
