Application value of LINC01600 in the clinical diagnosis of colorectal cancer
Objective To analyze the expression of long-chain non coding RNA LINC01600 in colorectal cancer(CRC)tissue and its value in the clinical diagnosis of CRC.Methods Clinical speci-mens of 37 patients with CRC admitted to our hospital from January 2020 to December 2023 were selected as research objects,and corresponding adjacent cancerous tissues were selected as controls.The total RNA was extracted from adjacent cancerous tissues and CRC tissues,and transcriptomic techniques were used to analyze the differentially expressed long chain non coding RNA between cancer tissues and adjacent cancer-ous tissues.The long-chain non coding RNA LINC01600 was selected as the research object.The expres-sion level of LINC01600 in CRC cancer tissues and adjacent tissues was detected by fluorescence quantita-tive polymerase chain reaction.A subject work characteristic curve was drawn to analyze the clinical diag-nostic efficacy of LINC01600 for CRC.The expression levels of LINC01600 in patients with different clini-cal stages and degrees of differentiation were analyzed by one-way ANOVA.The relationship between LINC01600 and patient prognosis was analyzed.Results A total of 39 123 long chain non coding RNAs were detected in CRC and adjacent tissues,including 2 018 new long chain non coding RNAs.Differential analysis showed that there were 1 626 differentially expressed long chain non coding RNAs,including 1 019 upregulated genes and 607 downregulated genes.Long chain non coding RNAs were mainly enriched in ribosomal pathways,citrate cycle pathways,and RNA splicing related pathways.The expression level of long chain non coding RNA LINC01600 in adjacent cancer tissues(0.85±0.10)was significantly lower than that in CRC tissues(1.99±0.31,t=21.390,P<0.05).The expression level of long chain non coding RNA LINC01600 in tumor tissues of medium to high differentiation patients(1.70±0.11)was sig-nificantly lower than that of low differentiation patients(2.19±0.24,t=7.510,P<0.05).The expres-sion level of long chain non coding RNA LINC01600 in CRC tissues of patients with lymph node metastasis(2.24±0.23)was significantly higher than that in CRC tissues(1.80±0.20,t=6.229,P<0.05).The expression level of long-chain non coding RNA LINC01600 in stage Ⅲ CRC tissues(2.19±0.32)was sig-nificantly higher than that in CRC tissues(1.92±0.27,t=2.796,P<0.05).The median survival time(14 months)of CRC patients with high expression of LINC01600 was significantly lower than that in pa-tients with low expression of LINC01600(26 months)(Log-rank=4.618,P<0.05).The area under curve(AUC)of long chain non coding RNA LINC01600 in tumor tissue for diagnosing CRC was 0.814[95%confidence interval(CI)=0.754-0.968,P<0.05].LINC01600 had a sensitivity of 92.45%and a specificity of 89.36%.Conclusion The level of long-chain non coding RNA LINC01600 in tumor tissue of CRC patients is significantly higher than that in adjacent tissues,and can be used as an auxiliary marker for preoperative and prognostic diagnosis in CRC.
