摘要

目的 观察青藤碱通过调节核因子E2相关因子2(Nrf2)/血红素氧合酶-1(HO-1)通路减少颅脑外伤引起的神经元凋亡.方法 采用随机数字表法将大鼠分为SIN组、SIN+Brusatol组和对照组,每组5只,建立模型后,实验组每天腹腔注射SIN[50 mg/(kg.d)];SIN+Brusatol组每天注射SIN[50 mg/(kg·d)]和Brusatol[1 mg/(kg·d)];对照组腹腔内注射等体积的生理盐水.蛋白质印迹法(Western blot)技术检测细胞中相关蛋白表达量;NSS评分量表评价青藤碱对神经功能影响;通过原位缺口末端标记法(TUNEL)染色,计算阳性细胞数量;定量资料比较采用t检验.结果 通过神经功能分析,SIN组NSS评分[(1.76±0.43)分]低于SIN+Brusatol组、对照组[(3.96± 0.45)、(4.02±0.49)分],差异有统计学意义(t=-10.76、-10.40,P<0.05).通过 TUNEL 染色统计阳性细胞数量,SIN组阳性细胞数量(52.33±5.13)明显低于SIN+Brusatol组、对照组(91.33± 7.10、99.33±5.86),差异有统计学意义(t=10.45、7.72,P<0.01).通过 Western blot 技术检测细胞中相关蛋白表达情况,SIN组Nrf2(1.35±0.05)表达量高于SIN+Brusatol组、对照组(0.81± 0.04、0.83±0.06),差异有统计学意义(t=24.47、20.17,P<0.05);SIN 组 HO-1(1.20±0.07)表达量高于 SIN+Brusatol 组、对照组(0.61±0.04、0.62±0.05),差异有统计学意义(t=20.84、22.30,P<0.05).结论 青藤碱通过调节Nrf2/HO-1通路减少颅脑外伤引起的神经元凋亡.

Abstract

Objective Sinomenine(SIN)was observed to reduce neuronal apoptosis induced by craniocerebral injury by regulating nuclear factor erythroid 2-related factor 2(Nrf2)/heme oxygenase-1(HO-1)pathway.Methods The rats were divided into SIN group,SIN+Brusatol group and control group by a random number table method.After establishing the model,the experimental group was intraperitoneally injected with SIN[(50 mg/(kg·d)].The SIN+Brusatol group was injected with SIN[50 mg/(kg·d)]and Brusatol[1 mg/(kg·d)].The control group was intraperitoneally injected with equal volume of normal sa-line.Western blotting was used to detect the expression of related proteins in cells.The NSS scale was used to evaluate the effect of SIN on neural function.The number of positive cells was calculated by termi-nal-deoxynucleotidyl transferase mediated nick end labeling(TUNEL)staining.SPSS 25.0 statistical soft-ware was used for analysis.Quantitative data were expressed as mean±standard deviation(x±s)and T-test was used.Results By neurological function analysis,the NSS score in the SIN group[(1.76± 0.43)points]was significantly lower than that in the SIN+Brusatol group and control group[(3.96± 0.45),(4.02±0.49)points,t=-10.76,-10.40,P<0.05].TUNEL staining showed that the num-ber of positive cells in the SIN group(52.33±5.13)was significantly less than that in the SIN+Brusatol group and control group(91.33±7.10,99.33±5.86,t=10.45,7.72,P<0.01).Western blotting showed that the expression of Nrf2(1.35±0.05)in the SIN group was significantly higher than that in the SIN+Brusatol group and control group(0.81±0.04,0.83±0.06,t=24.47,20.17,P<0.05).The expression of HO-1 in the SIN group(1.20±0.07)was significantly higher than that in the SIN+Brusatol group and control group(0.61±0.04,0.62±0.05,t=20.84,22.30,P<0.05).Conclusion SIN re-duces neuronal apoptosis induced by craniocerebral injury by regulating the Nrf2/HO-l pathway.
