Barrett食管组织中环氧合酶-2的表达及其与磷脂酰肌醇-3-激酶/蛋白激酶B/雷帕霉素靶蛋白通路的关系

摘要
目的 探究Barrett食管(BE)组织中磷脂酰肌醇-3-激酶(PI3K)/蛋白激酶B(Akt)/雷帕霉素靶蛋白(mTOR)信号通路与环氧合酶-2(COX-2)表达的关系及对Barrett食管发生的影响.方法 选择2021年1月至2023年1月间于山西医科大学第一医院治疗的BE患者共48例,取其中的病灶组织为BE组,同时取病灶周围正常食管黏膜组织标本作为对照组.对患者的BE组和对照组均进行食管组织免疫组织化学染色,检测组织中PI3K、Akt、mTOR以及COX-2蛋白的水平.应用Spearman秩相关方法,分别分析PI3K、Akt、mTOR与COX-2的相关性.结果 BE组的组织中COX-2蛋白阳性率及相对表达水平高于对照组(97.92%比70.83%、0.75±0.12比0.28±0.06,t=24.721,x2=85.429,P<0.05).BE组的组织中PI3K蛋白阳性率及相对表达水平高于对照组(85.42%比 11.63%、0.81±0.15 比 0.23±0.04,t=25.885,x2=91.132,P<0.05).BE 组的组织中PI3K蛋白阳性率及相对表达水平高于对照组(83.33%比14.58%、0.77±0.12比0.25±0.05,t=27.713,x2=89.273,P<0.05).BE组的组织中PI3K蛋白阳性率及相对表达水平高于对照组(81.25%比 33.33%、0.70±0.11 比 0.30±0.08,t=20.375,x2=75.422,P<0.05).BE 组的组织中 PI3K、Akt、mTOR 水平分别与 COX-2 水平呈正相关(r=0.681、0.605、0.634,P<0.05).结论 BE组织中PI3K/Akt/mTOR信号通路与COX-2水平呈正相关且表达水平上调,参与BE的发生发展.
Abstract
Objective To explore the association of phosphatidylinositol-3-kinase(PI3K)/protein kinase B(Akt)/mammalian target of rapamycin(mTOR)signaling pathway and cyclooxygenase-2(COX-2)expression in Barrett's esophagus(BE)and its influence on the development of Barrett's esophagus.Methods A total of 48 BE patients who were treated in the First Hospital of Shanxi Medical University from January 2021 to January 2023 were selected.The lesion tissue was selected as the BE group,and nor-mal esophageal mucosal tissue samples around the lesion were taken as the control group.The levels of PI3K,Akt,mTOR and COX-2 proteins in the BE group and control group were detected by immunohisto-chemical staining of esophageal tissues.Spearman rank correlation method was used to analyze the correla-tion between PI3K,Akt,mTOR and COX-2,respectively.Results The positive rate and relative expres-sion level of COX-2 protein in the BE group were higher than those in the control group(97.92%vs.70.83%;0.75±0.12 vs.0.28±0.06,t=24.721,85.429,P<0.05).The positive rate and relative expression level of PI3K in the BE group were higher than those in the control group(85.42%vs.11.63%;0.81±0.15 vs.0.23±0.04;t=25.885,91.132,P<0.05).The positive rate and relative expression level of PI3K in the BE group were higher than those in the control group(83.33%vs.14.58%;0.77±0.12 vs.0.25±0.05,t=27.713,89.273,P<0.05).The positive rate and relative expression level of PI3K in the BE group were higher than those in the control group(81.25%vs.33.33%;0.70±0.11 vs.0.30±0.08(t=20.375,75.422,P<0.05).In the BE group,PI3K,Akt and mTOR levels were positively correlated with COX-2 levels,respectively(r=0.681,0.605,0.634,P<0.05).Conclusion The PI3K/Akt/mTOR signaling pathway in BE tissues is positively correlated with the level of COX-2 and its expression level is up-regulated,which is involved in the occurrence and development of BE.
