Effect of ethanol on biophysical properties of red blood cells
Objective To study the effect of different doses of ethanol on the characteristics of red blood cells in blood.Methods A total of 8 normal blood samples were taken,each with 1 ml,one of which was for the control group without any treatment,and the other 6 were for the experimental group,and 20,40,60,80,100,120 μl ethanol were added,respectively,and then the electrophoretic rate of red blood cells under the influence of different concentrations of ethanol was observed.The changes of permea-bility brittleness and fluorescence spectra were compared between the ethanol group and the control group.Single factor analysis of variance was used for inter group data comparison.Results When the amount of ethanol added to the samples ranged from 0-40 μl,the electrophoretic rate gradually increased,and the electrophoretic rate of red blood cells added with 40 μl ethanol was the highest(1.192±0.280),which was significantly higher than that of the control group(0.803±0.180),and the difference was statistically significant(t=1.164,P<0.05).However,when the ethanol dose continued to increase,the electro-phoretic rate gradually decreased,and when the ethanol dose reached 80 μl,it was lower than the normal level.The electrophoretic rate of erythrocyte after adding ethanol 120 μl was significantly lower than the normal level(0.415±0.300)and control group(0.803±0.180),and the difference was statistically sig-nificant(t=0.418,P<0.05).When the amount of ethanol added to the sample was 40 μl,the permea-bility brittleness of the sample was(0.793±0.270),which was higher than that of the control group(0.674±0.540)and the difference was statistically significant(t=0.791,P<0.05).In order to excite the emission spectra of 407 nm and 532 nm,the peaks of the emission spectra appeared at 491 nm and 608 nm.From the point of view of the intensity of emission spectrum,when the amount of alcohol added was 5 μl,the intensity increased.The maximum strength reached when the amount of alcohol was 10 μl.When the equivalent reached 20 μl,its intensity decreased,which was less than the spectral intensity of red blood cell solution without alcohol.Conclusion Small dose of ethanol can promote the activity of e-rythrocyte membrane to some extent,but when the dose of ethanol is too large,the electrophoretic rate and osmotic fragility of red blood cells will decrease,affecting the structure of the erythrocyte membrane,and thus affecting the deformability of red blood cells and the activity of the membrane.
