Expression and clinical significance of regulators of G protein 16 in esophageal cancer
Objective To investigate the expression and clinical significance of regulators of G protein 16(RGS16)gene in esophageal cancer(ESCA).Methods The transcriptome data and clinical parameters from 160 cases of ESCA and 11 cases of adjacent cancers were downloaded from The Cancer Ge-nome Atlas(TCGA)database.The expression differences of RGS16 mRNA gene were analyzed by R lan-guage and Wilcoxon rank sum test.The correlation between RGS16 expression and clinical pathological characteristics was analyzed by Chi square test.The gene expression profiling interactive analysis(GEPIA)and multifactorial regression were used to analyze the prognostic factors of ESCA patients.RGS16 pathways were analyzed in R language to explore its potential mechanism in ESCA.Results The mRNA expression level of RGS16 in ESCA tissues was significantly higher than that in normal tissues[14.001(6.768,28.497)vs.3.088(2.133,5.560),Z=10.9,P<0.001],and its expression showed a positive correla-tion with the depth of tumour infiltration depth(x2=10.077,P<0.05).TCGA database analysis showed that patients with high RGS16 expression in ESCA tissues had a poor prognosis(Logrank P<0.05).The multifactorial analysis showed that RGS16 expression[hazard ratio=2.911,95%Confidence Interval:1.210-7.001,P<0.05]was an independent factor influencing the prognosis of ESCA.KEGG pathway a-nalysis showed that RGS16 expression was associated with phosphatidylinositol 3 kinase(PI3K)-protein ki-nase B(Akt)signaling pathway,adhesion plaques and other signaling pathways(P<0.05,q<0.05).Conclusion RGS16 gene is highly expressed in ESCA and is closely related to the poor prognosis of ESCA patients,and it may regulate the development of tumors through various signaling pathways,such as PI3K-Akt signaling pathway and Focal adhesion pathway.
