目的 探讨磷酸丝氨酸转氨酶1在食管癌组织中表达及对细胞增殖和耐药的影响.方法 收集郑州大学第一附属医院诊治的95例食管癌临床标本及癌旁组织作为研究对象,采用蛋白质免疫印迹分析癌旁组织和食管癌组织磷酸丝氨酸转氨酶1表达水平;人食管癌细胞系Ec-9706和顺铂耐药的Ec-9706随机分为对照短发卡RNA(shRNA)组和PSAT1 shRNA组,shRNA对照/耐药组和PSAT1 shRNA/耐药组,通过体内和体外细胞增殖实验分析磷酸丝氨酸转氨酶1对细胞体外增殖能力的影响;采用流式细胞术分析不同处理细胞的凋亡水平;采用蛋白质免疫印迹分析磷酸化磷脂酰肌醇3激酶(PI3K)、磷酸化蛋白激酶B(Akt)、耐药相关蛋白P-gp、剪切型半胱氨酰天冬氨酸特异性蛋白酶(Caspase)-3的表达水平.计量数据比较采用t检验.结果 癌旁组织中磷酸丝氨酸转氨酶1表达水平(0.91±0.20)明显低于食管癌组织表达水平(1.76±0.26),差异有统计学意义(t=25.230,P<0.05).对照shRNA组细胞抑制率[(7.14±2.06)%]明显低于PSAT1 shRNA组细胞[(54.79±11.97)%],差异有统计学意义(t=9.610,P<0.05).对照shRNA组细胞裸鼠成瘤体积和质量[(1 289.19±131.71)cm3、(5.36±0.67)g]高于 PSAT1 shRNA 组细胞[(705.66±65.88)cm3、(3.27±0.26)g],差异有统计学意义(t=9.706、7.110,P<0.05).shRNA对照/耐药组细胞抑制率[(21.80±3.39)%]明显低于PSAT1 shRNA/耐药组细胞[(41.12±3.59)%],差异有统计学意义(t=9.581,P<0.05).shRNA对照/耐药组细胞裸鼠成瘤体积和质量[(799.52±61.71)cm3、(3.68± 0.51)g]高于 PSAT1 shRNA/耐药组细胞[(558.62±44.97)cm3、(2.07±0.27)g],差异有统计学意义(t=7.728、6.932,P<0.05).对照 shRNA 组细胞抑制率[(3.78±0.81)%]明显低于 PSAT1 shRNA组细胞[(26.73±4.64)%],差异有统计学意义(t=11.940,P<0.05).shRNA对照/耐药组细胞抑制率[(16.75±4.95)%]明显低于PSAT1 shRNA/耐药组细胞[(35.04±4.05)%],差异有统计学意义(t=6.998,P<0.05).对照shRNA组细胞磷酸化PI3K、磷酸化Akt、耐药相关蛋白P-gp蛋白表达水平(0.91±0.10、1.26±0.18、1.19±0.12)明显高于 PSAT1 shRNA 组细胞(0.61±0.11、0.91±0.06、0.74±0.07),差异有统计学意义(t=5.024、4.487、7.693,P<0.05).结论 磷酸丝氨酸转氨酶1在食管癌组织中表达水平显著上调,参与食管癌细胞的增殖和耐药过程.
Expression of phosphoserine transaminase 1 in esophageal cancer tissue and its effects on cell prolif-eration and drug resistance
Objective To investigate the expression of phosphoserine aminotransferase 1 in esoph-ageal cancer tissue and its effects on cell proliferation and drug resistance.Methods Totally,95 cases of esophageal cancer clinical specimens and adjacent tissues were collected as the research objects.The ex-pression level of phosphoserine aminotransferase 1 in adjacent tissues and esophageal cancer tissues was an-alyzed by Western blotting.Human esophageal cancer cell line Ec-9706 and cisplatin-resistant Ec-9706 were randomly divided into control short hairpin RNA(shRNA)group and PSAT1 shRNA group,shRNA control/drug resistant group and PSAT1 shRNA/drug resistant group.The effect of phosphoserine amin-otransferase 1 on the proliferation ability of cells in vitro was analyzed by in vivo and in vitro cell prolifera-tion experiments.The apoptosis level of cells was analyzed by flow cytometry.The expression levels of phosphorylated phosphatidylinositol 3 kinase(PI3K),phosphorylated protein kinase B(Akt),drug resist-ance-related protein P-gp and spliced cysteinyl aspartate-specific protease(Caspase)-3 were analyzed by Western blotting.The t test was used for comparison of quantitative data.Results The expression level of phosphoserine aminotransferase 1 in adjacent tissues(0.91±0.20)was significantly lower than that in e-sophageal cancer tissues(1.76±0.26,t=25.230,P<0.05).The cell inhibition rate of control shRNA group[(7.14±2.06)%]was significantly lower than that of PSAT1 shRNA group[(54.79±11.97)%,t=9.610,P<0.05].The tumor volume and weight of nude mice in control shRNA group[(1 289.19± 131.71)cm3,(5.36±0.67)g]were greater than those in PSAT1 shRNA group[(705.66±65.88)cm3,(3.27±0.26)g,t=9.706,7.110,P<0.05].The inhibition rate of shRNA control/drug resistant group[(21.80±3.39)%]was significantly lower than that of PSAT1 shRNA/drug resistant group[(41.12± 3.59)%,t=9.581,P<0.05].The tumor volume and weight of nude mice in shRNA control/drug resist-ant group[(799.52±61.71)cm3,(3.68±0.51)g]were greater than those in PSAT1 shRNA/drug re-sistant group[(558.62±44.97)cm3,(2.07±0.27)g,t=7.728,6.932,P<0.05].The inhibition rate of control shRNA group[(3.78±0.81)%]was significantly lower than that of PSAT1 shRNA group[(26.73±4.64)%,t=11.940,P<0.05].The inhibition rate of shRNA control/drug resistant group[(16.75±4.95)%]was significantly lower than that of PSAT1 shRNA/drug resistant group[(35.04± 4.05)%,t=6.998,P<0.05].The protein expression levels of phosphorylated PI3K,phosphorylated Akt and drug resistance-related protein P-gp in control shRNA group(0.91±0.10,1.26±0.18,1.19± 0.12)were significantly higher than those in PSAT1 shRNA group(0.61±0.11,0.91±0.06,0.74± 0.07,t=5.024,4.487,7.693,P<0.05).Conclusion The expression level of phospho-serine amin-otransferase 1 in esophageal cancer tissues is significantly up-regulated,and it is involved in the prolifera-tion and drug resistance of esophageal cancer cells.
