Therapeutic effect of gastrodin on osteoarthritis in rats via Wnt/β-catenin pathway and nuclear factor-κB pathway
Objective To investigate the therapeutic effect of gastrodin on osteoarthritis in rats and the effects of gastrodin on Wnt/β-catenin pathway and nuclear factor-KB(NF-κB)pathway.Methods Totally,30 SD rats purchased from Henan Experimental Animal Center were divided into sham operation group,arthritis group and gastrodin group according to the random number table method,with 10 rats in each group.Knee osteoarthritis model was prepared by modified Hulth method in arthritis group and gastro-din group.The sham operation group underwent the same surgical procedure as the model group,but no os-teoarthritis model was formed.Gastrodin group rats were intraperitoneally injected with gastrodin(50 mg/kg),and the animals in sham operation group and arthritis group were intraperitoneally injected with equal volume of normal saline.After 8 weeks of treatment and anesthesia,knee cartilage tissues of three groups were taken for follow-up study.Hematoxylin-eosin staining was used to analyze the histopatho-logical changes of cartilage in the three groups.The levels of interleukin(IL)-1β,IL-6 and tumor necrosis factor α(TNFα)in articular cartilage of rats in the three groups were detected by enzymed-linked immu-nosorbent assay(ELISA).The expression levels of malondialdehyde and superoxide dismutase in articular cartilage were determined with malondialdehyde and superoxide dismutase kit.NF-κB,Toll-like receptor 4(TLR4)and the level of myeloid differentiation primary response protein 88(MyD88),type Ⅱ collagen,matrix metalloproteinase 13 and proteoglycan were analyzed by Western blotting in the three groups.The injury degree of articular cartilage in the three groups was analyzed by Mankin's score.One-way analysis of variance was used to compare the measurement data between groups.Results The levels of IL-1β,IL-6 and TNFα in articular cartilage of gastrodin group[(33.84±6.37),(21.64±3.98),(58.61± 8.83)pg/g]were lower than those in model group[(74.97±9.81),(45.63±7.87),(140.68± 20.79)pg/g,t=9.742,8.261,11.780,P<0.05].The level of malondialdehyde in articular cartilage of gastrodin group[(1.33±0.14)mmol/g]was significantly lower than that of model group[(2.38± 0.18)mmol/g,t=14.720,P<0.05].The activity of superoxide dismutase in articular cartilage of gast-rodin group[(86.29±6.99)U/mg]was significantly lower than that of model group[(65.17±7.37)U/mg,t=6.578.P<0.05].The levels of NF-κB,MyD88,Wnt2 and β-catenin in articular cartilage of gastrodin group(1.09±0.12,1.31±0.13,0.91±0.07,1.22±0.09)were significantly lower than those of model group(1.73±0.15,1.75±0.14,1.42±0.16,1.74±0.12,t=9.742,8.261,9.090,11.330,P<0.05).The levels of type Ⅱ collagen and proteoglycan in articular cartilage of gastrodin group(1.21±0.09,1.28±0.12)was significantly higher than in model group(0.77±0.10,0.87±0.11,t=10.240,7.971,P<0.05).The joint Mankin's score of gastrodin group(3.75±1.00)was significantly lower than that of model group(7.53±0.62,t=10.130,P<0.05).Conclusion Gastrodin can inhibit osteoarthritis in rats by inhibiting Wnt/β-catenin pathway and NF-κB pathway,delay articular cartilage degeneration,and achieve therapeutic effect on osteoarthritis.
