Comparative experimental study on muscle-derived stem cells and adipose-derived stem cells in the treatment of urethral sphincter dysfunction stress urinary incontinence in rats
Objective To investigate the effects and differences of muscle-derived stem cells(MD-SCs)and adipose-derived stem cells(ADSCs)in treating urethral sphincter dysfunction stress urinary in-continence(ISD)in rats.Methods MDSCs and ADSCs of rats were isolated and identified.After estab-lishing ray ISD model,the rats injected with MDSCs into bladder neck and external urethral sphincter served as MDSCs group(20 cases),and those injected with ADSCs served as ADSCs group(20 cases).The animals injected with D-Hank's culture solution served as control group(20 cases).The micturition function of three groups was evaluated by urodynamic detector before and 15,30 and 60 days after cell in-jection,respectively.Then the rats were killed.Results Urodynamic examination showed that as com-pared with the control group,both MDSCs injection therapy and ADSCs injection therapy improved the mic-turition function of ISD rats,and the therapeutic effect of MDSCs treatment group was more obvious.The maximum urinary pressure(MUP)in MDSCs group and ADSCs group was(16.95±3.80)and(12.71± 3.30),and the leak point pressure(LPP)was(58.49±7.60)and(45.61±8.80),which were signifi-cantly higher than in the control group(9.37±1.60,28.77±5.10).At 30th and 60th after the transplan-tation,the Maximum bladder capacity(MCC)in MDSCs group and ADSCs group decreased significantly as compared with control group(0.95±0.60 and 1.30±0.20 vs.1.86±0.90,P<0.05).Conclusion Injection of MDSCs and ADSCs has obvious effects oi the treatment of ISD,and efficacy of MDSCs injec-tion is better than ADSCs injection.
