Relationship between microRNA-141-3p,microRNA-223-3p levels in intestinal mucosal tissue and neonatal necrotizing small bowel colitis(NBC)condition and its predictive value
Objective To analyze the relationship between the levels of microRNA(miR)-141-3p and miR-223-3p in intestinal mucosal tissues and the condition of neonatal necrotizing enterocolitis(NEC)and its predictive value.Methods Totally,78 neonates with NEC admitted to Zhumadian Central Hospi-tal from 2021.1 to 2023.12 were selected as the observation group,and were divided into mild(n=22),moderate(n=30),and severe(n=26)subgroups according to the severity of the disease.In addition,80 cases of healthy newborns with physical examination results in the same time period were selected as the control group.miR-141-3p and miR-223-3p levels in the intestinal mucosa of the two groups were deter-mined by fluorescence quantitative polymerase chain reaction(PCR),and miR-141-3p and miR-223-3p levels in the observation group were compared between the children of the observation group with different disease severity,the comparison between two groups was conducted using t-test,while the comparison be-tween three groups was conducted using analysis of variance,and analyzed by Spearman's correlation anal-ysis.The relationship between miR-141-3p,miR-223-3p levels and the severity of disease in NEC neonates was analyzed.The predictive value of miR-141-3p,miR-223-3p levels on the condition of NEC neonates was assessed by plotting the receiving operative characteristics(ROC)curve and determining the area under the curve(AUC).Results The expression of miR-141-3p in the intestinal mucosal tissue of the observa-tion group was 1.65±0.38,while the expression of miR-141-3p in the intestinal mucosal tissue of the con-trol group was 0.94±0.20.The observation group was higher than the control group(t=14.748),and the differences between the groups were statistically significant(P<0.01).The expression of miR-223-3p in the intestinal mucosal tissue of the observation group was 0.75±0.14,while the expression of miR-223-3p in the intestinal mucosal tissue of the control group was 1.40±0.26.The expression of miR-223-3p in the observation group was lower than that in the control group(t=19.494),and the differences between the groups were statistically significant(P<0.01).The expression of miR-141-3p in the intestinal mucosal tis-sue of children in the mild,moderate,and severe groups was(1.29±0.21,1.60±0.25,1.92±0.34),while the expression of miR-223-3p in the intestinal mucosal tissue of children in the mild,moderate,and severe groups was(0.88±0.18,0.74±0.15,0.62±0.12).With the increase of disease severity in NEC neonates,the miR-141-3p level in their intestinal mucosal tissues showed a tendency to increase,and the miR-223-3p level showed a tendency to decrease,and there was statistically significant difference be-tween the groups(P<0.01).There was a positive correlation between miR-141-3p levels in intestinal mu-cosal tissues and the severity of NEC neonates(r=0.761,P<0.01),and a negative correlation between miR-223-3 p levels and the severity of NEC neonates(r=-0.655,P<0.01).miR-141-3p predicted whether the condition of NEC neonates was severe had an AUC of 0.947,a cut-off value of 1.650,a sensi-tivity of 0.923,and a specificity of 0.984.miR-223-3p predicted whether the condition of NEC neonates was severe had an AUC of 0.889,a cut-off value of 0.760,a sensitivity of 0.769,and a specificity of 0.885.Conclusion The relationship between miR-141-3p levels in intestinal mucosal tissue and the NEC neonates were positively correlated with the severity of their disease,and miR-223-3p level and the severity of their disease were negatively correlated,and both indexes had a high predictive value for the disease of NEC neonates.
