Expression of glycolysis-related enzymes and metabolites in glioma tissues and their correlation with immunosuppressive protein levels
Objective To investigate the expression of glycolysis-related enzymes and metabolites in glioma tissues and their correlation with immunosuppressive protein levels.Methods Glioma tissues and adjacent tissues obtained from our hospital from January 2022 to December 2023 were selected as the research objects.The expression levels of glycolysis-related enzymes in adjacent tissues and glioma tissues were analyzed by fluorescence quantitative polymerase chain reaction(PCR).The changes of pyruvate and lactic acid were measured by a kit.The infiltration levels of CD8 positive T lymphocytes in adjacent tissues and glioma tissues were analyzed by immunofluorescence.The expression levels of programmed death ligand-1 protein in adjacent tissues and glioma tissues were analyzed by immunohistochemistry.The t test was used for comparison of measurement data between groups.Results The mRNA expression levels of hexokinase 2(HK2),phosphoglycerate kinase 1(PGK1),lactate dehydrogenase A(LDHA)and pyruvate kinase 2(PKM2)in glioma tissues(1.53±0.19,1.70±0.22,1.81±0.18,1.79±0.22)were signifi-cantly higher than those in adjacent tissues(0.81±0.16,1.01±0.17,0.88±0.14,0.98±0.18,t=24.950,19.180,27.900,18.850,P<0.05).The levels of pyruvate and lactic acid in glioma tissues[(56.24±14.27),(8.31±1.96)mg/ml protein]were significantly higher than those in adjacent tissues[(29.87±11.29),(2.84±1.02)mg/ml protein,t=9.721,16.570,P<0.05].The proportion of CD8 lymphocytes in glioma tissues[(2.48±0.59)%]was significantly lower than that in adjacent tissues[(5.06±1.17)%,t=13.160,P<0.05].The expression levels of programmed death ligand-1 and pro-grammed death receptor-1 in glioma tissues(1.57±0.19,1.62±0.19)were significantly higher than those in adjacent tissues(0.72±0.12,0.87±0.14,t=16.3900,13.770,P<0.05).The mRNA ex-pression levels of hexokinase 2,phosphoglycerate kinase 1,lactate dehydrogenase A and pyruvate kinase 2 were positively correlated with the protein expression level of programmed death ligand-1(r=0.145,0.105,0.217,0.209,P<0.05).Conclusion The glycolytic enzymes hexokinase 2,phosphoglycerate kinase 1,lactate dehydrogenase A and pyruvate kinase are significantly increased in glioma tissues,and are positively correlated with the expression level of programmed death ligand-1.
