Apoptosis of gastric mucosa cells and expression of epidermal growth factor receptor,neurofila-ment protein 200,human epithelial growth factor receptor-2 protein in precancerous rats
Objective To investigate the apoptosis of gastric mucosa cells and the expression of epidermal growth factor receptor(EGFR),neurofilament protein 200(NF-200)and human epithelial growth factor receptor-2(C-ERB-2)in rat model of gastric precancerous lesion.Methods Totally,60 healthy Wistar rats were randomly divided into model and control groups,with 30 animals in each group.Model group was treated with methylnnitroguanidine for 90 days to establish a rat model of pregastric cancer.Control group was treated with the same dose of distilled water of gavage.The apoptosis rate of gas-tric epithelial cells and the protein expression levels of EGFR,NF-200 and C-ERB-2 in gastric mucosa of rats in the two groups were analyzed.Pearson correlation analysis was performed.In addition,the efficacy of EGFR,NF-200 and C-ERB-2 proteins in diagnosis of gastric precancerous lesions was determined by the receiver operating characteristic(ROC)curve analysis.Group t test was used to compare the measurement data between groups.Results The apoptosis rate of gastric mucosal epithelial cells in model group was(37.22±0.49)%,which was higher than in control group[(1.12±0.03)%,P<0.05].The expression levels of EGFR and C-ERB-2 in gastric mucosa of the model group were(278.31±32.84)and(86.03±7.15),which were higher than control group[(181.37±21.34),(33.05±5.32)],while the expression level of NF-200 was(0.71±0.03),which was lower than control group(1.00±0.00,all P<0.05).Pearson correlation analysis showed apoptosis rate of gastric mucosa cells was positively correlated with the protein expression levels of EGFR and C-ERB-2,and negatively correlated with the protein expression level of NF-200(all P<0.05).Conclusion Gastric mucosal cells in rat model of gastric precancerous lesions have obvious apoptosis,and the expression of EGFR and C-ERB-2 proteins is significantly increased,and the expression of NF-200 protein is significantly decreased.The combined detection of the expression of the a-bove three proteins is helpful for the early diagnosis of gastric precancerous lesions.
