Expression of triple domain protein 59 and nuclear factor inhibitory protein in gastric cancer tissue and their relationship with chemotherapy efficacy and prognosis in patients
Objective To detect the expression of tripartite motif-containing protein 59(TRIM59)and nuclear factor inhibitor protein(IκBα)in gastric cancer tissues,and to analyze their relationship with the efficacy of chemotherapy and prognosis in patients.Methods A total of 103 patients with gastric canc-er who underwent surgical treatment from January 2020 to January 2023 were selected,and the adjacent tis-sues resected more than 2 cm away from the cancerous tissues were selected as controls.The expression of TRIM59 and IκBα in gastric cancer tissues and adjacent tissues was compared.Pearson correlation analysis was used to analyze the correlation between TRIM59 and IκBα expression in gastric cancer tissues.All pa-tients were divided into drug-resistant group and sensitive group according to the efficacy of chemotherapy after 3 cycles of treatment.The general data and the expression of TRIM59 and IκBα in gastric cancer tis-sues were compared between the drug-resistant group and the sensitive group.Logistic regression model was used to analyze the influencing factors of the efficacy of gastric cancer chemotherapy.Kaplan-Meier curve was drawn to analyze the relationship between the expression of TRIM59 and IκBα in gastric cancer tissues and the progression and survival of gastric cancer.Count data is represented as[n(%)]and tested using a chi square test;The K-S test is used to represent econometric data that conforms to a normal distribution as mean±standard deviation((x)±s),and independent sample t-test is used for inter group comparison.Results Immunofluorescence experiments showed that TRIM59 and IκBα could be detected in the same cellular regions with two different fluorescence signals,representing co-localization in the nucleus.Com-pared with adjacent tissues,the fluorescence intensity of TRIM59 and IκBα in gastric cancer tissues was increased(P<0.05).The expression level of TRIM59 in gastric cancer tissue was higher than that in ad-jacent tissues(17.39±4.04 vs.8.35±2.05,t=20.251,P<0.01).The expression level of IκBα in gastric cancer tissue was higher than that in adjacent tissues(15.40±3.82 vs.7.16±1.97,t=19.457,P<0.01).Pearson correlation analysis showed that the expression of TRIM59 and IκBα in gastric cancer tissues was positively correlated(r=0.341,P<0.01).Logistic regression analysis showed that age,TNM stage Ⅲ/Ⅳ,undifferentiated/low differentiation,TRIM59 expression,and IκBα expression were influen-cing factors of the efficacy of chemotherapy in patients with gastric cancer(P<0.05).The expression level of TRIM59 in drug-resistant gastric cancer tissue was higher than that in sensitive group tissue(19.35±3.47 vs.14.42±2.96,t=7.473,P<0.01).The expression level of TRIM59 in drug-resistant gastric cancer tissue was higher than that in sensitive group tissue(17.21±3.19 vs.12.65±3.04,t=7.234,P<0.01).Among the 103 patients with gastric cancer,15 were lost to follow-up,and the remaining 88 were followed up for 8-39 months.The patients were divided into high TRIM59 expression group and high IκBα expression group if the expression was higher than the median value,and into low TRIM59 expression group and low IκBα expression group if the expression was lower than the median value.The median pro-gression-free survival(PFS)of high TRIM59 expression was shorter than that of low TRIM59 expression[24(22,25.5)months vs.39(28,50)months,x2=6.885,P<0.05].The PFS with high IκBα ex-pression was shorter than that with low IκBα expression[24(17.5,30.5)months vs.33(20,39)months,x2=7.145,P<0.05].The median overall survival(OS)of high TRIM59 expression was shorter than that of low TRIM59 expression[33(20,46)months vs.38(36,40)months,x2=4.340,P<0.05].The OS with high IκBα expression was shorter than that with low IκBα expression[30(19.5,40.5)months vs.38(36,40)months,x2=7.283,P<0.05].Conclusion The expression of TRIM59 and Iκ Bα in gastric cancer tissues increases,and the expression levels are positively correlated.Both of them are closely related to the efficacy of gastric cancer chemotherapy and prognosis.
