Expression of transcription factor E2F1 in pituitary tumor and its effect on glucose metabolism in pituitary tumor cells
Objective To investigate the expression of transcription factor E2F1 in pituitary tumors and its effect on glucose metabolism in pituitary tumors.Methods From January 2021 to December 2023,102 cases of pituitary tumor tissues and para-cancerous tissues treated in the First Affiliated Hospital of Xinxiang Medical University were selected as the study objects.The expression levels of transcription factor E2F1 protein and mRNA in para-cancerous tissue and pituitary tumor tissue were analyzed by Western blot-ting and quantitative fluorescent PCR.Human pituitary tumor cell line AtT20 was randomly divided into control group and E2F1 KD group.Control short hairpin RNA(shRNA)and E2F1 shRNA lentivirus infec-ted cells were used to establish stable cell lines.Cell counting kit-8(CCK-8)and clonal formation assays were used to analyze cell proliferation ability in control group and E2F1 KD group.The protein and mRNA expression levels of key glycolytic enzymes(hexokinase,6-phosphofructokinase,pyruvate kinase and lactate dehydrogenase)in tumor and cell lines were analyzed by Western blotting and quantitative fluores-cent PCR.The levels of lactate,pyruvate and Phosphoenolpyruvate in cell lines were determined by a kit.T test was used to compare measurement data between groups.Results The expression level of E2F1 pro-tein in pituitary tumor tissues(1.53±0.17)was significantly higher than that in adjacent tissues(0.70±0.15,t=37.420,P<0.05).The absorbance(A)value of the control group(1.86±0.06)was signifi-cantly higher than that of the E2F1 KD group(1.40±0.11,t=9.169,P<0.05).The rate of cell clonal formation in control group[(68.57±9.87)%]was significantly higher than that in E2F1 KD group[(33.53±3.99)%,t=8.059,P<0.05].The expression levels of hexokinase,6-phosphofructokinase,pyruvate kinase and lactate dehydrogenase(1.31±0.08,0.98±0.10,1.27±0.11,0.96±0.06)were significantly higher than E2F1 KD cells(0.88±0.07,0.52±0.14,0.71±0.14,0.64±0.06,t=10.250,6.486,7.878,9.001,P<0.05).The mRNA expression levels of hexokinase,6-phosphofruc-tokinase,pyruvate kinase and lactate dehydrogenase in control group(1.05±0.07,1.11±0.09,0.93±0.10,1.06±0.09)were significantly higher than those in E2F1 KD group(0.68±0.05,0.73±0.07,0.65±0.11,0.62±0.10,t=10.070,8.091,4.728,7.887,P<0.05).The relative concentrations of lactic acid,pyruvate and phosphoenolpyruvate(21.83±3.43,17.33±2.94,22.33±2.42)were signifi-cantly higher than E2F1 KD group(15.33±1.75,12.83±1.47,15.50±1.87,t=4.134,3.349,5.469,P<0.05).Conclusion The expression level of transcription factor E2F1 is significantly increased in pituitary tumor tissues,and it is involved in the proliferation of pituitary tumor cells by regulating the ex-pression of glycolysis gene.
