Expression of WW domain-containing oxidoreductase in intracranial aneurysms and as a molecular marker
Objective At present,there is no clear molecular markers for early screening of in-tracranial aneurysms(IA).The value of WW domain-containing oxidoreductase(WWOX)as an IA screen-ing indicator was analyzed.Methods Through a case-control study,102 IA patients(41 males and 61 fe-males;78 unruptured and 24 ruptured)and 100 healthy people(59 males and 61 females)and 20 tissue samples(8 cases of IA tissue,12 cases of superficial temporal artery)collected from the First Affiliated Hospital of Xinjiang Medical University from 2021 to 2023 were subjected to real-time fluorescence quanti-tative polymerase chain reaction(RT-qPCR)and immunofluorescence staining.Enzyme linked immunosor-bent assay(ELISA)was performed on 42 plasma samples(8 males and 13 females in the IA group;10 males and 11 females in the control group),and ROC curve modeling was performed after multi-factor Logistic regression analysis.The relationship between WWOX genes and IA was analyzed.The independ-ent sample t test was used for comparison between two groups,and one-way analysis of variance was used for comparison between multiple groups.Results The WWOX gene content in tissues of IA group was lower than that of the control group(15.100±12.751 vs.0.788±0.598,t=3.16,P<0.01),and immu-nofluorescence staining showed that WWOX levels were reduced in IA group.The blood WWOX gene con-tent in the IA group was higher than in the healthy people(0.715±1.647 vs.1.484±0.971,t=7.524,P<0.01).The blood WWOX gene content in the control group was lower than in the unruptured and rup-tured groups(0.701±0.152 vs.1.434±0.745 vs.1.005±0.597,F=40.52,P<0.01).The blood WWOX gene content in the female IA group was higher than that in the male IA group(0.924±0.476 vs.1.601±0.916,t=3.845,P<0.01).ELISA showed that the WWOX protein content in the IA group was higher than that in the control group(148.651±18.071 vs.169.366±25.998,t=2.642,P<0.05).The blood and plasma ROC curve results showed that the curves shifted to the upper left(AUC=0.762 4,error 0.035 73;AUC=0.727 9,error 0.088 67).Conclusion WWOX is involved in the occurrence and de-velopment of IA,and the WWOX gene in the blood can be used as a molecular marker for early clinical screening of IA.
