Expression of long non-coding RNA NORAD in superficial esophageal squamous cell carcinoma and its clinical significance
Objective To investigate the expression of long non-coding RNA(lncRNA)in superfi-cial esophageal squamous cell carcinoma and its clinical significance.Methods A total of 54 cases of superficial esophageal cancer and adjacent tissues treated from June 2022 to June 2023 were selected as re-search objects in Xinxiang Central Hospital and Shanghai Chest Hospital.LncRNA microarray was used to analyze lncRNA differentially expressed in adjacent tissues and tumor tissues,and the functional enrich-ment and biological regulatory pathways of differentially expressed genes were analyzed.LncRNA NORAD with the most significant difference was selected as the study objects,and the expression level of lncRNA NORAD in adjacent and superficial esophageal cancer tissues was quantitatively analyzed by fluorescence.The relationship between lncRNA NORAD expression level and clinicopathological features and prognosis of patients with superficial esophageal cancer was analyzed.Multivariate regression analysis was used to ana-lyze the independent prognostic factors of superficial esophageal cancer.Measurement data were compared by t test.Results In this study,a total of 2 180 differentially expressed lncRNAs were screened using ln-cRNA chips(1 432 lncRNAs up-regulated and 748 lncRNAs down-regulated).LncRNA NORAD had the largest up-regulation ratio(12 times).Kyoto encyclopedia of genes and genomes(KEGG)enrichment showed that these lncRNAs were involved in many biological metabolic and signal transduction related path-ways,including cellular glucose metabolism,cell growth and death,protein interactions,protein transcrip-tional post-translational modification,DNA damage repair,and amino acid purine metabolism.The expres-sion level of lncRNA NORAD in adjacent tissues(1.01±0.17)was significantly lower than that in superfi-cial tumor cells(1.95±0.17,t=29.150,P<0.05).LncRNA NORAD expression level in superficial tumor tissues of medium-high differentiation patients(1.85±0.12)was significantly lower than that of low-differentiation patients(2.07±0.15,t=5.828,P<0.05).The expression level of lncRNA NORAD(2.03±0.14)in superficial tumor tissues of patients with bureaucratic stenosis was significantly higher than that of patients without bureaucratic stenosis(1.88±0.16).The difference was statistically significant(t=3.592,P<0.05).The expression level of lncRNA NORAD in superficial tumor tissues of patients with venous cancer thrombolus(2.05±0.17)was significantly higher than that of patients without venous cancer thrombolus(1.90±0.13,t=5.248,P<0.05).The expression level of lncRNA NORAD in superficial tumor tissues of patients with lymph node metastasis(1.83±0.13)was significantly higher than that of pa-tients without lymph node metastasis(2.04±0.13,t=6.221,P<0.05).LncRNA NORAD high expres-sion,differentiation degree and lymph node metastasis were independent risk factors for prognosis of superfi-cial esophageal cancer(Wald=14.526,13.578,11.425,P<0.05).Conclusion LncRNA NORAD is highly expressed in superficial esophageal cancer,and is closely related to the clinicopathological features and prognosis of patients.
