Relationship between the expression of Dickkopf related protein 3 and ubiquitin-binding enzyme E2T in bladder cancer tissue and paracancer tissue with clinicopathological features and prognosis
Objective To investigate the relationship between the expression of Dickkopf related protein 3(DKK3)and ubiquitin-binding enzyme E2T(UBE2T)in bladder cancer tissues and the clinico-pathological features and prognosis of patients.Methods A total of 73 patients with bladder cancer who were diagnosed in China Resources WISCOM General Hospital from February 2019 to March 2021 were se-lected.The bladder cancer tissues were taken as the study group,and adjacent tissues as the control group.Spearman rank correlation was used to analyze the correlation between DKK3 and UBE2T in bladder cancer tissue.Cox regression analysis was used to analyze the factors affecting the prognosis of bladder cancer patients.Kaplan-Meierwas used to analyze the relationship between DKK3,UBE2T with the progno-sis of bladder cancer patients.Results The negative expression rate of DKK3 and the positive expression rate of UBE2T in bladder cancer tissues were higher than those in adjacent tissues(73.97%vs.50.68%,69.86%vs.21.92%,x2=8.430,33.790,P<0.05).DKK3 and UBE2T were negatively correlated in bladder cancer tissue,with correlation coefficient r=-0.631,P<0.01.The proportions of DKK3 nega-tive expression and UBE2T positive expression in bladder cancer patients with tumor diameter>2 cm(66.67%,66.67%),TNM stage T2-T4(70.37%,68.63%),and lymph node metastasis(68.52%,74.51%)were higher than those in patients with tumor diameter ≤2 cm(33.33%,33.33%),TNM stage Ta-T1(29.63%,31.37%),and non lymph node metastasis(31.48%,25.49%)(x2=5.777,4.932,11.848,P<0.05).Tumor diameter[hazard ratio(HR)=3.750],TNM stage(HR=1.972),lymph node metastasis(HR=1.166),and UBE2T(HR=4.679)were prognostic risk factors for patients with bladder cancer(P<0.05),and DKK3(HR=0.857)was a prognostic protective factor for patients with bladder cancer(P<0.05).There were 54 patients with negative expression of DKK3 who survived 31,19 patients with positive expression of DKK3 who survived 17,and survival was higher in patients with DKK3-positive expression than in DKK3-negative patients(89.47%vs.57.41%,x2=5.073,P<0.05).There were 51 patients with positive expression of UBE2T who survived 29,and 22 patients with negative expression of UBE2T who survived 19.Survival was higher in UBE2T-negative expressing patients than in UBE2T-positive patients(86.36%vs.56.86%,x2=4.702,P<0.05).Conclusion The ex-pression of DKK3 and UBE2T in bladder cancer is related to the severity of clinicopathological features and closely related to the prognosis of bladder cancer patients.
