Expression and clinical significance of small nuclear ribonucleo protein polypeptide E in prostate cancer
Objective To investigate the expression of small nuclear ribonucleo protein polypep-tide E(SNRPE)in prostate cancer(PCa)and its regulatory function in prostate cancer cells.Methods Based on the cancer genome atlas(TCGA)database,the differential analysis,survival analysis and clini-cal correlation analysis of SNRPE gene were performed.Two groups of short interfering RNA(siRNA)were used to knock down the expression of SNRPE,and the effects on the proliferation,migration and invasion of prostate cancer cells were investigated.Independent sample t test was used for comparison between different groups.Results The expression level of SNRPE in prostate cancer samples was significantly high-er than that in normal prostate samples(5.3±0.10 vs.4.8±0.12,t=5.54,P<0.01).The expression of SNRPE in the T1+T2 stage was significantly lower than that in the T3+T4 stage expression group(t=4.53,P<0.05).The expression of SNRPE1 in patients with prostate cancer without lymph node metasta-sis was significantly lower than that in patients with lymph node metastasis(t=4.21,P<0.05).Multiva-riable Cox regression analysis showed that:age[hazard ratio(HR):0.99(0.95-1.00),P>0.05],T stage[HR:1.84(0.96-3.50),P>0.05]and N stage[HR:1.32(0.68-2.60),P>0.05]cannot be used as an independent prognostic factor related to OS,while SNRPE expression[HR:2.00(1.20-3.50),P<0.05]and Gleason score[HR:3.14(1.57-6.30),P<0.01]can be used as a prognostic factor in prostate cancer.In PC3,the wound healing area of ctrl group was significantly higher than that of the si-SNRPE#1 and si-SNRPE#2 groups,and the difference between the groups was statistically significant[(82.30±1.21)%vs.(38.10±2.56)%,t=27.04,P<0.01;(82.30±1.21)%vs.(40.05±1.82)%,t=33.48,P<0.01].Furthermore,the number of migration cells in ctrl,si-SNRPE#1 and si-SNRPE#2 PC3 groups were 351.8±16.5,189.4±10.50,176.6±23.5 respectively,and the difference was significant statistically(t=14.38,10.57,P<0.01).Moreover,the number of invasion cells in ctrl,si-SNRPE#1 and si-SNRPE#2 groups were 495.6±77.9,251.2±70.8 and 203.4±41.2 respectively,and the difference was significant statistically(t=4.02,5.74,P<0.01).Conclusion SNRPE gene can promote the proliferation,migration and invasion of prostate cancer cells.SNRPE is expected to be a new biomarker for the diagnosis and targeted therapy of prostate cancer.
