Expression of microRNA-151a-3p in non-small cell lung cancer tissues and its relationship with radiotherapy sensitivity
Objective To investigate the expression of microRNA(miR)-151a-3p in non-small cell lung cancer(NSCLC)and its relationship with radiotherapy sensitivity.Methods Totally,54 patients with NSCLC admitted to our hospital from December 2019 to August 2023 were selected as the study objects.All patients underwent three-dimensional conformal radiotherapy,and the radiotherapy efficacy of the patients was evaluated.The efficacy of patients was fallen into the following categories:complete re-sponse,partial response,stable and progressive.The patients obtaining the complete remission(n=22)and partial remission(n=15)served as the effective treatment group(n=37),and those obtaining stable response(n=10)and progressive response(n=7)as ineffective treatment group(n=17).The expression level of miR-151a-3p in tumor tissues in the effective treatment group and the ineffective treatment group was analyzed by fluorescence quantitative polymerase chain reaction(PCR).According to the mean expres-sion level of miR-151a-3p,the patients were divided into miR-151a-3p high expression group and miR-151a-3p low expression group.The risk factors leading to lung cancer radiotherapy sensitivity were an-alyzed by multi-factor regression to explore the correlation between the expression level of miR-151a-3p and radiotherapy resistance of lung cancer.Independent samples t test was used to compare measurement data between groups.Results 22 were in complete remission,15 were in partial remission,10 were stable and 7 were progressing.A total of 37 patients with complete and partial responses were included in the effective treatment group,and 17 patients with stable and progressive responses were included in the ineffective treatment group.The effective rate of radiotherapy was 62.58%(37/54).The expression level of miR-151a-3p in tumor tissues of the effective treatment group(0.94±0.17)was significantly lower than that of the ineffective treatment group(1.49±0.29,t=8.807,P<0.05).The effective rate of three-di-mensional conformal intensity modulated radiotherapy in patients with poorly differentiated tumors[87.5%(21/24)]was significantly higher than that in radiotherapy for medium to high differentiated tumors[53.33%(16/30),x2=8.694,P<0.05].The effective rate of three-dimensional conformal intensity modulated radiotherapy in patients with lymph node metastasis[54.29%(19/35)]was significantly lower than that in patients without lymph node metastasis[94.74%(18/19),x2=8.694,P<0.05].The effec-tive rate of three-dimensional conformal intensity modulated radiotherapy in patients with low expression of miR-151a-3p[75.00%(30/40)]was significantly higher than that in patients with high expression of miR-151a-3p[50.00%(7/14),x2=8.694,P<0.05].Multivariate logistic regression analysis showed that low differentiation,no lymph node metastasis and low expression level of miR-151a-3p were predictive factors for the sensitivity of three-dimensional conformal intensity modulated radiotherapy in NSCLC[odds ratio(OR)=3.412,3.876,2.985,P<0.05].Correlation analysis showed that the expression level of miR-151a-3p in NSCLC tissue was negatively correlated with the sensitivity of three-dimensional conformal intensity modulated radiotherapy(r=-0.479,P<0.05).Conclusion MiR-151a-3p in tumor tissue of lung cancer patients is closely related to radiotherapy sensitivity,which can be used to evaluate clinical ra-diotherapy effect.
