Expression of cholinergic receptor nicotinic α5 in prostate adenocarcinoma and its effect on the metastatic activity of prostate adenocarcinoma cells
Objective To investigate the expression of cholinergic receptor nicotinic alpha 5(CHRNA5)in prostate cancer and its effect on the malignant metastasis of prostate cancer DU145 cells.Methods Prostate cancer tissues and adjacent normal prostate tissues were obtained from 15 prostate ade-nocarcinoma patients who underwent surgery in Tianjin Medical University General Hospital from January,2022 to October,2023.Immunohistochemistry was used to detect the expression level of CHRNA5 in pros-tate cancer tissues and adjacent normal prostate tissues.DU145 cells were divided into three experimental groups:si CHRNA5 group,si NC group and yes-associated protein 1/Transcriptional coactivator with PDZ-binding motif(YAP1/TAZ)inhibitor group.The proliferation of DU145 cells was detected by methyl-thiazolyl-tetrazolium(MTT)assay.The invasion ability of DU145 cells was analyzed by Transwell assay.The migration ability of DU145 cells was analyzed by cell scratch test.The apoptosis rate of DU145 cells was analyzed by flow cytometry.The expression levels of YAP1/TAZ signaling pathway and CHRNA5 pro-tein were detected by Western blotting.Comparison between the two groups was done using independent sample t test.Results The expression level of CHRNA5 in prostate cancer tissues was higher than that in adjacent normal prostate tissues(0.28±0.07 vs.0.89±0.08,t=12.178,P<0.05).The proliferation ability of DU145 cells in si CHRNA5 and YAP1/TAZ inhibitor groups was lower than that in si NC group(1.72±0.06 vs.0.83±0.01 vs.0.81±0.05,F=11.369,P<0.05).The invasion number of DU 145 cells in the si CHRNA5 and YAP1/TAZ inhibitor groups was less than that in the si NC group(139.19±10.76 vs.83.89±9.36 vs.79.93±8.25,F=21.328,P<0.05).The migration rate of DU 145 cells in the si CHRNA5 and YAP1/TAZ inhibitor groups was lower than that in the si NC group[(88.65±6.06)%vs.(55.86±5.33)%vs.(56.23±3.88)%,F=20.162,P<0.05].The apoptosis rate of DU145 cells in the si CHRNA5 and YAP1/TAZ inhibitor groups was higher than that in the si NC group[(3.16±0.36)%vs.(12.62±0.25)%vs.(13.26±0.17)%,F=13.179,P<0.05].The protein levels of YAP1 and TAZ in DU145 cells of si CHRNA5 group and YAP1/TAZ inhibitor group were lower than those of si NC group(0.81±0.12 vs.0.32±0.15 vs.0.28±0.09,F=15.782,P<0.05;0.79±0.12 vs.0.18±0.09 vs.0.22±0.06,F=17.336,P<0.05).The expression levels of YAP1 and TAZ protein in pcDNA3.1 CHRNA5 group were higher than those in pcDNA3.1 NC group(0.32±0.06 vs.0.78±0.03,t=11.569,P<0.05;0.26±0.07vs.0.72±0.03,t=16.238,P<0.05).Conclusion CHRNA5 is highly expressed in prostate cancer tissues.CHRNA5 can promote the proliferation,migration and invasion of prostate cancer DU145 cells by activating the YAP1/TAZ pathway.
