Objective To investigate the effects of microRNA-27a(miR-27a)on proliferation and migration of diffuse large B-cell lymphoma cells by regulating the expression of B-cell translocation gene 1(BTG1).Methods Human U2932 cells were randomly divided into control group,miR-27a mimic nega-tive control group(mimic NC group),miR-27a mimic group(miR-27a mimic group)and miR-27a inhibi-tor negative control group(inhibitor NC group)and miR-27a inhibitor group(miR-27a inhibitor group),according to random number table.Control group was replaced with fresh medium,and other groups were transfected with mimic NC,miR-27a mimic,inhibitor NC and miR-27a inhibitor,respectively.After trans-fection 24 h,the expression of miR-27a was detected by real-time fluorescence quantitative reverse tran-scription polymerase chain reaction(qRT-PCR),cell proliferation activity was detected by methyl thiazolyl tetrazolium(MTT)assay,the number of migrating cells was detected by Transwell assay,and the protein expression levels of BTG1,epithelial cadherin,neural cadherin and vimentin were detected by western blot.One-way analysis of variance was used for comparison between groups.Results The expression of miR-27a,cell proliferation activity and number of migrated cells of miR-27a mimic group were higher than those of mimic NC group[2.53±0.07 vs.1.03±0.04,(117.25±3.10)%vs.(98.25±3.44)%,(38.46±3.22)cells vs.(23.95±6.79)cells,t=32.225,7.107,3.344,all P<0.05];The expres-sion of miR-27a,cell proliferation activity and number of migrated cells of miR-27a inhibitor group were lower than those of miR-27a inhibitor group[0.54±0.03 vs.1.02±0.06,(71.25±3.73)%vs.(98.62±2.84)%,(11.25±3.37)cells vs.(21.88±4.41)cells,t=12.394,10.112,3.317,allP<0.05];The expressions of BTG1 and epithelial cadherin protein in miR-27a mimic group were lower than those of mimic NC group(0.16±0.03 vs.0.41±0.05,0.09±0.01 vs.0.44±0.06,t=7.426,9.966,all P<0.05).The expressions of neural cadherin protein and Vimentin(0.65±0.06 vs.0.38±0.07,0.74±0.06 vs.0.31±0.04,t=5.072,10.328,all P<0.05)were higher than those of mimic NC group.The expression levels of BTG1 and epithelial cadherin protein in miR-27a inhibitor group were higher than those in inhibitor NC group(0.86±0.08 vs.0.38±0.07,0.71±0.06 vs.0.42±0.05,t=7.821,6.431,all P<0.05).The expression levels of neural cadherin protein and Vimentin were lower than those of inhibitor NC group(0.08±0.01 vs.0.37±0.06,0.12±0.03 vs.0.35±0.07,t=8.258,5.231,all P<0.05).Conclusion Down-regulation of miR-27a targeting BTG1 can inhibit the prolifera-tion and migration of diffuse large B-cell lymphoma cells and play an anti-tumor role.
