Effects of microRNA-27a on proliferation and migration of U2932 cells

目的 探讨微小RNA-27a(miR-27a)通过调控B细胞易位基因1(BTG1)表达对人弥漫大B细胞淋巴瘤细胞增殖和迁移的影响.方法 人源性U2932细胞按照随机法分为对照组、miR-27a模拟物阴性对照组(mimic NC组)、miR-27a模拟物组(miR-27a mimic组)、miR-27a抑制剂阴性对照组(inhibitor NC组)和miR-27a抑制剂组(miR-27a inhibitor组),对照组细胞更换新鲜培养基,其余组分别转染 mimic NC、miR-27a mimic、inhibitor NC 和 miR-27a inhibitor,转染 24 h 后实时荧光定量反转录聚合酶链反应(qRT-PCR)法检测miR-27a表达量,噻唑蓝(MTT)法检测细胞增殖活性,Transwell实验检测迁移细胞数,蛋白印迹法检测BTG1、上皮钙黏蛋白、神经钙黏蛋白和波形蛋白表达量.组间比较采用单因素方差分析(Oneway-ANOVA).结果 miR-27a mimic组miR-27a表达量、细胞增殖活性和迁移细胞数均高于mimic NC组[2.53±0.07比1.03±0.04、(117.25±3.10)%比(98.25±3.44)%、(38.46±3.22)个 比(23.95±6.79)个,t=32.225、7.107、3.344,P均<0.05];miR-27a inhibitor组miR-27a表达量、细胞增殖活性和迁移细胞数均低于inhibitor NC组[0.54±0.03 比 1.02±0.06、(71.25±3.73)%比(98.62±2.84)%、(11.25±3.37)个 比(21.88±4.41)个,t=12.394、10.112、3.317,P均<0.05];miR-27a mimic 组 BTG1 和上皮钙黏蛋白表达量均低于 mimic NC 组(0.16±0.03 比 0.41±0.05、0.09±0.01 比 0.44±0.06,t=7.426、9.966,P均<0.05),神经钙黏蛋白和波形蛋白表达量均高于mimic NC组(0.65±0.06比0.38±0.07、0.74±0.06 比 0.31±0.04,t=5.072、10.328.P 均<0.05);miR-27a inhibitor组BTG1 和上皮钙黏蛋白表达量均高于 inhibitor NC 组(0.86±0.08 比 0.38±0.07、0.71±0.06 比 0.42±0.05,t=7.821、6.431,P均<0.05),神经钙黏蛋白和波形蛋白表达量均低于inhibitor NC组(0.08±0.01比0.37±0.06、0.12±0.03 比 0.35±0.07,t=8.258、5.231,P 均<0.05).结论 下调 miR-27a 靶向调控BTG1,可抑制弥漫大B细胞淋巴瘤细胞增殖和迁移,发挥抗肿瘤作用.
Objective To investigate the effects of microRNA-27a(miR-27a)on proliferation and migration of diffuse large B-cell lymphoma cells by regulating the expression of B-cell translocation gene 1(BTG1).Methods Human U2932 cells were randomly divided into control group,miR-27a mimic nega-tive control group(mimic NC group),miR-27a mimic group(miR-27a mimic group)and miR-27a inhibi-tor negative control group(inhibitor NC group)and miR-27a inhibitor group(miR-27a inhibitor group),according to random number table.Control group was replaced with fresh medium,and other groups were transfected with mimic NC,miR-27a mimic,inhibitor NC and miR-27a inhibitor,respectively.After trans-fection 24 h,the expression of miR-27a was detected by real-time fluorescence quantitative reverse tran-scription polymerase chain reaction(qRT-PCR),cell proliferation activity was detected by methyl thiazolyl tetrazolium(MTT)assay,the number of migrating cells was detected by Transwell assay,and the protein expression levels of BTG1,epithelial cadherin,neural cadherin and vimentin were detected by western blot.One-way analysis of variance was used for comparison between groups.Results The expression of miR-27a,cell proliferation activity and number of migrated cells of miR-27a mimic group were higher than those of mimic NC group[2.53±0.07 vs.1.03±0.04,(117.25±3.10)%vs.(98.25±3.44)%,(38.46±3.22)cells vs.(23.95±6.79)cells,t=32.225,7.107,3.344,all P<0.05];The expres-sion of miR-27a,cell proliferation activity and number of migrated cells of miR-27a inhibitor group were lower than those of miR-27a inhibitor group[0.54±0.03 vs.1.02±0.06,(71.25±3.73)%vs.(98.62±2.84)%,(11.25±3.37)cells vs.(21.88±4.41)cells,t=12.394,10.112,3.317,allP<0.05];The expressions of BTG1 and epithelial cadherin protein in miR-27a mimic group were lower than those of mimic NC group(0.16±0.03 vs.0.41±0.05,0.09±0.01 vs.0.44±0.06,t=7.426,9.966,all P<0.05).The expressions of neural cadherin protein and Vimentin(0.65±0.06 vs.0.38±0.07,0.74±0.06 vs.0.31±0.04,t=5.072,10.328,all P<0.05)were higher than those of mimic NC group.The expression levels of BTG1 and epithelial cadherin protein in miR-27a inhibitor group were higher than those in inhibitor NC group(0.86±0.08 vs.0.38±0.07,0.71±0.06 vs.0.42±0.05,t=7.821,6.431,all P<0.05).The expression levels of neural cadherin protein and Vimentin were lower than those of inhibitor NC group(0.08±0.01 vs.0.37±0.06,0.12±0.03 vs.0.35±0.07,t=8.258,5.231,all P<0.05).Conclusion Down-regulation of miR-27a targeting BTG1 can inhibit the prolifera-tion and migration of diffuse large B-cell lymphoma cells and play an anti-tumor role.
