摘要
目的 探讨LINC00963在胃癌组织中的表达变化及其与胃癌患者临床病理特征和预后的关系.方法 选取我院胃癌根治术患者127例,手术后收集胃癌组织和癌旁组织,采用荧光定量聚合酶链反应(PCR)分析LINC00963表达水平.采用免疫组织化学分析层粘连蛋白γ1(LAMC1)、黏着斑激酶(FAK)和波形蛋白(Vimentin)表达阳性率;分析LINC00963表达水平与患者临床病理特征、预后以及LAMC1、FAK和Vimentin阳性率的相关性.组间比较采用独立样本t检验.结果 癌旁组织LINC00963表达水平(1.01±0.16)明显低于胃癌组织(1.59±0.17),差异有统计学意义(t=29.660,P＜0.05).LAMC1、FAK 和 Vimentin 阳性率分别为 42.52％(54/127)、52.76％(67/127)和 55.91％(71/127).LINC00963 表达水平与 LAMC1、FAK 和 Vimentin 阳性率呈正相关(r=3.124、2.978、3.784,P＜0.05).低分化患者肿瘤组织LINC00963表达水平(1.75±0.10)明显高于中高分化患者(1.52±0.14),差异有统计学意义(t=8.892,P＜0.05).淋巴结转移患者肿瘤组织LINC00963表达水平(1.72±0.09)明显高于中高分化患者(1.47±0.14),差异有统计学意义(t=11.730,P＜0.05).Ⅲ～Ⅳ期患者肿瘤组织LINC00963表达水平(1.70±0.10)明显高于Ⅰ～Ⅱ期患者(1.50±0.12),差异有统计学意义(t=9.885,P＜0.05).预后不良组患者肿瘤组织LINC00963表达水平(1.77±0.08)明显高于预后良好者(1.47±0.09),差异有统计学意义(t=9.885,P＜0.05).胃癌患者淋巴结转移、TNM分期、细胞分化程度、LINC00963表达水平是导致胃癌患者死亡的危险因素(P＜0.05).结论 LINC00963在胃癌组织中表达水平明显增加,与LAMC1、FAK和Vimentin阳性率呈正相关,与患者临床分期、分化程度和淋巴结转移密切相关,其表达水平越高,患者预后越差.
Abstract
Objective To investigate the expression of LINC00963 in gastric cancer and its rela-tionship with clinicopathological features and prognosis of gastric cancer patients.Methods A total of 127 patients with gastric cancer undergoing radical gastrectomy in our hospital were selected.The gastric cancer tissues and adjacent tissues were collected after surgery,and the expression level of LINC00963 was analyzed by fluorescence quantitative polymerase chain reaction(PCR).The positive rates of laminin γ1(LAMC1),focal adhesion kinase(FAK)and Vimentin were analyzed by immunohistochemistry.The cor-relation between the expression level of LINC00963 and the clinicopathological characteristics,prognosis and the positive rates of lamc1,FAK and Vimentin were analyzed.The measurement data between groups were compared by independent sample t test.Results The expression level of LINC00963 in adjacent tis-sues(1.01±0.16)was significantly lower than that in gastric cancer tissues(1.59±0.17,t=29.660,P＜0.05).The positive rates of lamc1,FAK and Vimentin were 42.52％(54/127),52.76％(67/127)and 55.91％(71/127),respectively.The expression level of LINC00963 was significantly correlated with the positive rates of lamc1,FAK and Vimentin(r=3.124,2.978,3.784,P＜0.05).The expression level of LINC00963 in tumor tissues of poorly differentiated patients(1.75±0.10)was significantly higher than that of moderately well differentiated patients(1.52±0.14,t=8.892,P＜0.05).The expression level of l LINC00963 in tumor tissues of patients with lymph node metastasis(1.72±0.09)was signifi-cantly higher than that of moderately well differentiated patients(1.47±0.14,t=11.730,P＜0 the ex-pression level of LINC00963 in tumor tissue of patients with stage Ⅲ-Ⅳ(1.70±0.10)was significantly higher than that of patients with stage Ⅰ-Ⅱ(1.50±0.12,t=9.885,P＜0.05).The expression level of LINC00963 in tumor tissue of patients with poor prognosis group(1.77±0.08)was significantly higher than that of patients with good prognosis group(1.47±0.09,t=9.885,P＜0.05).Lymph node metasta-sis TNM stage,cell differentiation and LINC00963 expression level were the risk factors of death in patients with gastric cancer(P＜0.05).Conclusion The expression level of LINC00963 in gastric cancer tissue is significantly increased,which is signiifcantly correlated with the positive rate of LAMC1,FAK and Vim-entin,and is closely related to the clinical stage,differentiation and lymph node metastasis of patients.The higher the expression level,the worse the prognosis of patients.
