Expression and clinical value of cytokine signaling inhibitor 1 in nasopharyngeal carcinoma
Objective To explore the expression and value of cytokine signaling inhibitor 1 in na-sopharyngeal carcinoma.Methods A total of 69 cases of nasopharyngeal carcinoma tissues and corre-sponding adjacent tissues from our hospital from June 2019 to December 2023 selected as the research sub-jects.The expression level of cytokine signaling inhibitor 1 was analyzed by Western blotting and fluores-cence quantitative PCR.The relationship between the expression level of inhibitor of cytokine signaling pro-tein 1 and the positive rate of Ki-67 and PCNA in nasopharyngeal carcinoma tissues was analyzed.The di-agnostic value of the expression level of inhibitor of cytokine signaling protein 1 and the clinicopathological characteristics and prognosis of patients with nasopharyngeal carcinoma was analyzed.The measurement da-ta between groups were compared by independent sample t test.Results The expression levels of inhibitor of cytokine signaling protein 1 mRNA and protein in nasopharyngeal carcinoma tissues(0.64±0.15,0.54±0.16)were significantly lower than those in adjacent tissues(1.04±0.14,0.93±0.19,t=15.830,12.710,P<0.05).The expression level of inhibitor of cytokine signaling protein 1 in Ki-67 and PCNA positive patients(0.48±0.12,0.47±0.12)was significantly lower than that in nasopharyngeal carcinoma tissues of Ki-67 and PCNA positive patients(0.74±0.09,0.70±0.11,t=7.531,7.310,P<0.05).The expression level of inhibitor of cytokine signaling protein 1 in nasopharyngeal carcinoma tissues of pa-tients with undifferentiated carcinoma(0.46±0.15)was significantly lower than that of patients with dif-ferentiated non keratinizing carcinoma(0.59±0.13,t=3.579,P<0.05).The expression level of inhib-itor of cytokine signaling protein 1 in nasopharyngeal carcinoma tissues of patients with poorly differentiated carcinoma(0.44±0.14)was significantly lower than that of patients with moderately well differentiated carcinoma(0.63±0.11,t=5.216,P<0.05).The expression level of inhibitor of cytokine signaling pro-tein 1 in nasopharyngeal carcinoma tissues of stage Ⅲ-Ⅳ patients(0.43±0.13)was significantly lower than that of patients with medium to high differentiation(0.62±0.12,t=4.876,P<0.05).The survival time of patients in the high expression group[(20.36±4.71)months]was significantly higher than that of patients in the low expression group[(15.80±4.59)months,log rank=6.201,P<0.05].The area un-der the curve of inhibitory protein of cytokine signaling 1 predicting the poor prognosis of nasopharyngeal carcinoma was 0.845.The specificity was 75.63%,and the sensitivity was 81.54%.High TNM stage and low expression of inhibitor of cytokine signaling protein 1 were independent risk factors for poor prognosis of nasopharyngeal carcinoma(P<0.05).Conclusion The low expression of inhibitor of cytokine signaling protein 1 in nasopharyngeal carcinoma is the main factor of poor prognosis of nasopharyngeal carcinoma.
