摘要
铁死亡是一种以铁积累和脂质过氧化为特征的铁依赖性程序性细胞死亡形式.铁死亡细胞中存在线粒体活性氧(ROS)超载、铁超载、三羧酸循环(TAC)及线粒体DNA(mtDNA)应激、内质网(ER)应激、溶酶体自噬、溶酶体铁和一氧化氮积累、高尔基体应激、核糖体蛋白合成速率减慢等变化,这些病理性或应激性变化亦可影响铁死亡的发生发展.本文拟综述近五年细胞器特异性变化影响铁死亡的机制,为铁死亡介导的疾病防治提供新思路.
Abstract
Ferroptosis is a form of iron-dependent programmed cell death characterized by iron accumulation and lipid peroxidation.Changes such as mitochondrial reactive oxygen species(ROS)overload,iron overload,tricar-boxylic acid cycle(TAC)and mitochondrial DNA(mtDNA)stress,endoplasmic reticulum(ER)stress,lysosomal au-tophagy,lysosomal iron and nitric oxide accumulation,Golgi stress,and ribosomal protein synthesis rate are present in ferroptotic cells.These pathological or stressful changes can also affect the occurrence and development of ferroptosis.This paper reviews the mechanism of organelle specific changes affecting ferroptosis in recent five years,and provides new ideas for the prevention and treatment of ferroptosis-mediated diseases.
