Experimental study of human umbilical cord mesenchymal stem cell conditioned medium for treat-ment of rat interstitial cystitis
Objective To investigate the efficacy of human umbilical cord mesenchymal stem cell conditioned media(HUC-MSC-CM)bladder perfusion in rat interstitial cystitis(IC).Methods 30 adult SD female rats were randomly averaged into three groups by random digital table method,the IC and MSC-CM groups constructed a model of interstitial cystitis using cyclophosphamide intraperitoneal injection,the blank group was intraperitoneally injected with an equal volume of normal saline;The MSC-CM group underwent HUC-MSC-CM bladder perfusion therapy after successful modeling,every 3 days for 1 week,the blank group and the IC group were treated with equal volume normal saline bladder perfusion;after treat-ment,the urination interval,concentration of bladder inflammatory factor[interleukin(IL)-1 β,IL-6,tumor necrosis factor-α(TNF-α)],degree of pathological inflammation,mast cell count,and apoptotic cell count were analyzed and compared.The independent sample t test was used for comparison between groups.Results The urination interral in the MSC-CM group was larger than that in the IC group[(254.40±4.75)s vs.(191.90±4.95)s,t=9.111,P<0.05],the concentration of bladder homoge-nization of inflammatory factors(IL-1β,IL-6,TNF-α)was lower in the MSC-CM group than that in the IC group[(116.80±8.34)pg/ml vs.(202.00±10.79)pg/ml,(177.20±54.22)pg/ml vs.(283.80±42.86)pg/ml,(51.46±8.68)pg/ml vs.(70.43±14.53)pg/ml,t=6.251,4.876,3.545,P<0.05],and the pathological score,mast cell count,and apoptotic cell count were lower in the MSC-CM group than those in the IC group[(9.20±0.49)cells vs.(12.40±0.52)cells,(7.50±0.65)cells vs.(11.50±0.54)cells,(17.00±1.23)cells vs.(28.80±2.13)cells,t=4.476,4.707,4.793,P<0.05)].Conclusion Human umbilical cord mesenchymal stem cells conditioned medium can effectively treat interstitial cystitis in rats.
