目的 探讨蛋白质精氨酸甲基转移酶5在肝癌中的表达与肝癌辐射敏感性的关系.方法 选取2022年1月至2024年1月我院收治放疗的30例肝癌患者作为研究对象,采用荧光定量聚合酶链反应(PCR)和蛋白质印迹法(Western blot)分析肝癌和癌旁组织中蛋白质精氨酸甲基转移酶5表达水平.根据两组患者放疗敏感性分为敏感组(15例)和耐受组(15例),分析两组患者肝癌组织蛋白质精氨酸甲基转移酶5表达水平.SMMC7721人肝癌细胞随机分为对照组和敲低组,采用蛋白质精氨酸甲基转移酶5短发卡RNA(shRNA)和对照shRNA慢病毒感染两组细胞,建立蛋白质精氨酸甲基转移酶5敲低和对照细胞系.两组细胞分别采用6 MV-X线(6Gy)照射细胞,采用细胞计数试剂盒(CCK-8)实验分析细胞的增殖能力;采用细胞凋亡检测试剂盒测定细胞的凋亡能力;建立移植瘤裸鼠模型,放疗治疗后,测定肿瘤体积,采用原位缺口末端标记法(TUNEL)染色分析小鼠肿瘤组织细胞凋亡情况.组间比较采用独立样本t检验.结果 癌旁组织中蛋白质精氨酸甲基转移酶5 mRNA和蛋白质表达水平(0.82±0.20、0.47±0.19)低于肝癌组织(1.63±0.16、1.32±0.27),差异有统计学意义(t=23.110、19.610,P<0.05).放疗敏感组患者肝癌组织蛋白质精氨酸甲基转移酶5 mRNA和蛋白质表达水平(1.54±0.11、1.12±0.11)低于耐受组(1.80±0.11、1.33±0.24),差异有统计学意义(t=9.318、7.853,P<0.05).对照组细胞CCK-8吸光度值(1.60±0.12)高于敲低(1.19±0.09),差异有统计学意义(t=10.120,P<0.05).对照组细胞凋亡比例[(25.85±3.57)%]低于敲低组[(56.45±3.42)%],差异有统计学意义(t=10.120,P<0.05).对照组细胞裸鼠成瘤体积[(978.62±77.85)mm3]高于敲低组[(528.89±108.29)mm3],差异有统计学意义(t=9.537,P<0.05).对照组裸鼠肿瘤细胞TUNEL阳性率[(18.72±1.71)%]低于敲低组[(56.45±3.42)%],差异有统计学意义(t=27.950,P<0.05).对照组细胞c-Met和Jab1/CSN5 mRNA表达水平(0.97±0.08、0.92±0.06)高于敲低组(0.62±0.08、0.46±0.12),差异有统计学意义(t=9.318、7.853,P<0.05).结论 放疗耐受患者蛋白质精氨酸甲基转移酶5表达水平显著增加,通过介导c-Met和Jab1/CSN5表达水平降低肝癌放疗敏感性.
Relationship between the expression of protein arginine methyltransferase 5 and radiation sensitivity of hepatocellular carcinoma
Objective To investigate the relationship between the expression of protein arginine methyltransferase 5 and radiation sensitivity of liver cancer.Methods 30 patients with liver cancer treated with radiotherapy were selected as the study objects From January 2022 to January 2024.The expression level of protein arginine methyltransferase 5 in liver cancer and adjacent tissues was analyzed by fluores-cence quantitative polymerase chain reaction(PCR)and Western blotting.The two groups were divided in-to sensitive group and tolerant group according to radiotherapy sensitivity.The expression level of protein arginine methyltransferase 5 in liver cancer tissues of the two groups was analyzed.SMMC7721 human hep-atocellular carcinoma cells were randomly divided into control group and knockdown group.The two groups of cells were infected with arginine methyltransferase 5 short hairpin RNA(shRNA)and control shRNA lentivirus,and the protein arginine methyltransferase 5 knockdown and control cell lines were established.The cells in the two groups were irradiated with 6 MV-X(6 Gy),and the proliferation ability of the cells was analyzed by cell counting kit-8(CCK-8).The apoptotic capacity of the cells was measured by apopto-sis detection kit.The nude mouse model of transplanted tumor was established,the tumor volume was measured after radiotherapy,and the apoptosis of tumor tissue was analyzed by terminal-deoxynucleotidyl transferase mediated nick end labeling(TUNEL)staining.Independent sample t test was used to compare the measurement data between groups.Results The expression levels of protein arginine methyltransferase 5 mRNA and protein in adjacent tissues(0.82±0.20,0.47±0.19)were significantly lower than those in liver cancer tissues(1.63±0.16,1.32±0.27,t=23.110,19.610,P<0.05).The protein arginine methyltransferase 5 mRNA and protein expression levels in the radiotherapy sensitive group(1.54±0.11,1.12±0.11)were significantly lower than those in the tolerant group(1.80±0.11,1.33±0.24,t=9.318,7.853,P<0.05).The absorbance value of CCK-8 in the control group(1.60±0.12)was signifi-cantly higher than that in the knockdown group(1.19±0.09,t=10.120,P<0.05).The proportion of apoptosis in the control group[(25.85±3.57)%]was significantly lower than that in the knockdown group[(56.45±3.42)%,t=10.120,P<0.05].The tumor formation volume of the control group[(978.62±77.85)mm3]was significantly higher than that of the knockdown Group[(528.89±108.29)mm3,t=9.537,P<0.05].The TUNEL positive rate of tumor cells in the control group[(18.72±1.71)%]was significantly lower than that in the knockdown group[(56.45±3.42)%,t=27.950,P<0.05].The mRNA expression levels of c-Met and Jab1/CSN5 in the control group(0.97±0.08,0.92±0.06)were significantly higher than those in the knockdown group(0.62±0.08,0.46±0.12,t=9.318,7.853,P<0.05).Conclusion The expression level of protein arginine methyltransferase 5 is significant-ly increased in patients with radiotherapy tolerance,which can reduce the radiotherapy sensitivity of HCC by mediating the expression level of c-Met and Jab1/CSN5.
