Relationship between the expression of protein arginine methyltransferase 5 and radiation sensitivity of hepatocellular carcinoma
Objective To investigate the relationship between the expression of protein arginine methyltransferase 5 and radiation sensitivity of liver cancer.Methods 30 patients with liver cancer treated with radiotherapy were selected as the study objects From January 2022 to January 2024.The expression level of protein arginine methyltransferase 5 in liver cancer and adjacent tissues was analyzed by fluores-cence quantitative polymerase chain reaction(PCR)and Western blotting.The two groups were divided in-to sensitive group and tolerant group according to radiotherapy sensitivity.The expression level of protein arginine methyltransferase 5 in liver cancer tissues of the two groups was analyzed.SMMC7721 human hep-atocellular carcinoma cells were randomly divided into control group and knockdown group.The two groups of cells were infected with arginine methyltransferase 5 short hairpin RNA(shRNA)and control shRNA lentivirus,and the protein arginine methyltransferase 5 knockdown and control cell lines were established.The cells in the two groups were irradiated with 6 MV-X(6 Gy),and the proliferation ability of the cells was analyzed by cell counting kit-8(CCK-8).The apoptotic capacity of the cells was measured by apopto-sis detection kit.The nude mouse model of transplanted tumor was established,the tumor volume was measured after radiotherapy,and the apoptosis of tumor tissue was analyzed by terminal-deoxynucleotidyl transferase mediated nick end labeling(TUNEL)staining.Independent sample t test was used to compare the measurement data between groups.Results The expression levels of protein arginine methyltransferase 5 mRNA and protein in adjacent tissues(0.82±0.20,0.47±0.19)were significantly lower than those in liver cancer tissues(1.63±0.16,1.32±0.27,t=23.110,19.610,P<0.05).The protein arginine methyltransferase 5 mRNA and protein expression levels in the radiotherapy sensitive group(1.54±0.11,1.12±0.11)were significantly lower than those in the tolerant group(1.80±0.11,1.33±0.24,t=9.318,7.853,P<0.05).The absorbance value of CCK-8 in the control group(1.60±0.12)was signifi-cantly higher than that in the knockdown group(1.19±0.09,t=10.120,P<0.05).The proportion of apoptosis in the control group[(25.85±3.57)%]was significantly lower than that in the knockdown group[(56.45±3.42)%,t=10.120,P<0.05].The tumor formation volume of the control group[(978.62±77.85)mm3]was significantly higher than that of the knockdown Group[(528.89±108.29)mm3,t=9.537,P<0.05].The TUNEL positive rate of tumor cells in the control group[(18.72±1.71)%]was significantly lower than that in the knockdown group[(56.45±3.42)%,t=27.950,P<0.05].The mRNA expression levels of c-Met and Jab1/CSN5 in the control group(0.97±0.08,0.92±0.06)were significantly higher than those in the knockdown group(0.62±0.08,0.46±0.12,t=9.318,7.853,P<0.05).Conclusion The expression level of protein arginine methyltransferase 5 is significant-ly increased in patients with radiotherapy tolerance,which can reduce the radiotherapy sensitivity of HCC by mediating the expression level of c-Met and Jab1/CSN5.
