Expression of CXC chemokine receptor 4 in thyroid cancer and its relationship with prognosis and immune invasion
Objective To investigate the expression of C-X-C chemokine receptor type 4(CXCR4)in thyroid cancer and its relationship with prognosis and immune invasion.Methods 98 cases of thyroid cancer and adjacent tissues treated from January 2020 to January 2022 in Yantai Mountain Hospi-tal of Shandong were selected as the study objects,and CXCR4 mRNA and protein expression levels in ad-jacent tissues and thyroid cancer tissues were analyzed by fluorescent quantitative polymerase chain reaction(PCR)and Western blotting.Oncomine database,Kaplan_meier Plotter database and TIMER database were used to analyze the relationship between CXCR4 gene expression and immune cell invasion and prog-nosis of thyroid cancer.Independent sample t test was used to compare the measurement data between groups.Results The mRNA expression level of CXCR4 in para-carcinoma tissues(1.05±0.20)was sig-nificantly lower than that in thyroid cancer tissues(2.00±0.22,t=31.480,P<0.05).The mRNA ex-pression level of CXCR4 in paracancer tissues(0.69±0.25)was significantly lower than that in thyroid cancer tissues(1.45±0.24,t=21.550,P<0.05).The results of a large number of analyses in the data-base showed that the mRNA expression level of CXCR4 in adjacent tissues(1.57±0.52)was significantly lower than that in thyroid cancer tissues(3.41±0.68),and the difference was statistically significant(t=8.914,P<0.05).CXCR4 gene expression was correlated with the proportion of activated natural killer cells and M0 macrophages(r=0.498,0.399,P<0.05).CXCR4 gene expression was negatively correla-ted with the proportion of resting state natural killer cells,resting state memory CD4 T lymphocytes,resting mast cells and monocytes(r=-1.524,-1.948,-3.102,-2.145,P<0.05).The overall survival time of thyroid cancer patients with high CXCR4 expression[(12.52±3.14)months]was significantly lower than that of thyroid cancer patients with low CXCR4 expression[(22.41±4.56)months],and the difference was statistically significant(Log-Rank=3.108,P<0.05).Conclusion The expression of CXCR4 mRNA and protein is significantly increased in thyroid cancer tissues,and is closely related to im-mune cell infiltration in patients.The higher the expression of CXCR4,the worse the prognosis of patients.
