Expression and clinical significance of the CCAAT enhancer-binding protein β in gastric cancer tissues
Objective To observe the expression of the CCAAT enhancer-binding protein β(C/EBPβ)in gastric cancer tissues and explore its clinical significance.Methods From January 2021 to December 2023,the tumors and their paired adjacent normal tissues from 100 gastric cancer patients were collected in Zhejiang Cancer Hospital,and the expression levels of C/EBPβ protein and mRNA in the specimens were measured by immunohistochemistry and real-time RT-PCR,and the correlation between C/EBPβ expression and the clinicopathological parameters and the impact on patient prognosis were ana-lyzed.Measurement data were analyzed by Person x2 test.Results The high expression rate of C/EBPβprotein and mRNA were 59.0%(59/100)and 69.0%(69/100),respectively,which were significantly higher than those in the adjacent normal tissues(13.0%,13/100;3.0%,3/100).The expression level of C/EBPβ protein in gastric cancer tissues was correlated with tumor size(67.74%vs.42.11%),tumor site(distal vs.cardia)(52.87%vs.92.31%,the depth of infiltration(10.53%vs.69.14%),lymphat-icvessel invasion(0.00%vs.61.70%),lymph node metastasis(0.00%vs.68.24%),distant metasta-sis(55.79%vs.100.00%)and TNM stage(4.17%vs.75.00%)(x2=11.833,4.665,21.653,9.140,9.140,3.690,39.328,P<0.05);but had no significant correlation with gender(59.72%vs.53.57%),age at tumor diagnosis(65.52%vs.54.93%),tumour differentiation(56.36%vs.60.00%)and venous invasion(52.63%vs.61.29%)(x2=0.395,0.940,0.134,0.792,P>0.05).Correlation analysis between C/EBPβ mRNA expression levels in GC tissues and clinicopathological param-eters of patients showed the same trend.The expression level of C/EBPβ protein in gastric cancer tissue was not significantly related to patient prognosis,there was no significant difference in median OS between them both(median OS:15 months vs.15 months)(x2=48.439,P<0.05).The expression level of C/EBPβ mRNA in gastric cancer tissue was not significantly related to patient prognosis,there was no sig-nificant difference in median OS between them both(median OS:16 months vs.16 months)(x2=53.175,P<0.05).Cox univariate regression analysis showed that the depth of tumor invasion(HR=2.054,95%CI:1.188-3.552),lymph node metastasis(HR=2.408,95%CI:0.966-6.003),distant metastasis(HR=2.663,95%CI:1.063-6.671)and TNM stage(HR=2.344,95%CI:1.394-3.941)were sig-nificantly associated with the prognosis of gastric cancer patients(P<0.05),but Cox multivariate regres-sion analysis found that the depth of tumor invasion(HR=1.404,95%CI:0.691-2.855),lymph node metastasis(HR=1.518,95%CI:0.542-4.256),distant metastasis(HR=1.867,95%CI:0.715-4.875)and TNM stage(HR=1.642,95%CI:0.803-3.355)were independent prognostic factors of gas-tric cancer patients(P>0.05).Conclusion C/EBPβ is highly expressed in gastric cancer tissues and promotes gastric cancer progression,but it does not affect patient prognosis.
