Study on the expression of SRY related high mobility group box 7 in colon cancer patients and its correlation with pathological characteristics
Objective To explore the expression of SRY related high mobility group box 7(SOX7)in colon cancer and its correlation with clinicopathology and prognosis through bioinformatics,so as to pro-vide reference for clinical diagnosis,treatment and prognosis improvement of colon cancer patients.Methods A total of 524 cases of colon cancer patients with RNA sequencing downloaded from the cancer genome atlas(TCGA)were included,Organize pathological data such as patient survival time,tumor size,clinical stage,survival status,differentiation degree,lymph node metastasis,gender,and age.There were 475 patients without missing values,including 40 normal samples and 435 tumor samples.The DESeq2 package of R software was used to analyze 475 differentially expressed genes(DEGs)between normal and tumor tissues in the TCGA database.Kaplan Meier survival analysis was applied to compare the survival rates of patients in the high expression and low expression groups.The overall survival rate of patients was used as a measure to plot the survival curve.Kyoto encyclopedia of genes and genomes(KEGG),gene set enrichment analysis(GSEA),and gene ontology(GO)pathway enrichment analysis of SOX7 gene.The comparison between groups was conducted using t test or x2 test.Results Using the TCGA database to an-alyze differentially expressed genes between normal and cancer tissues of colon cancer,6 531 stable genes,2 310 down regulated genes,and 2 218 up regulated genes were identified.In the TCGA dataset,a com-parison of 40 paired samples showed that the expression level of SOX7 in colorectal cancer tissues was sig-nificantly reduced compared to normal tissue(x2=15.607,P<0.05).There was no statistically significant difference in the expression level of SOX7 among patients in terms of age,tumor size,gender,TNM stage,nerve invasion,and vascular invasion(x2=1.669,0.276,3.108,0.055,0.630,1.906,P<0.05);The expression level of SOX7 showed statistically significant differences in lymph node metastasis,differen-tiation degree,and infiltration depth among patients(x2=10.918,8.948,33.642,P<0.05).compared with the high expression group of SOX7,the overall survival rate of patients in the low expression group was significantly reduced(x2=12.325,P<0.05).Conclusion SOX7 is lowly expressed in colon cancer tis-sues and is significantly correlated with the clinical pathological characteristics and prognosis of patients.
