摘要

目的 探讨葡萄糖苷木糖基转移酶2(GXYLT2)在胃癌中的表达及其对患者预后的影响,并分析其与肿瘤浸润免疫细胞的相关性以及对胃癌细胞生物学特性的影响.方法 基于癌症基因组图谱(TCGA)数据库中的数据,将胃癌患者按GXYLT2表达中位数分为高/低表达组,采用独立样本t检验以及卡方检验分析GXYLT2在胃癌组织中的表达及其与患者生存的关系.通过单变量和多变量Cox比例风险回归分析评估GXYLT2与临床特征的预后相关性.体外实验将HGC-27胃癌细胞分为Control GXYLT2组(转染阴性对照siRNA)和si-GXYLT2组(转染siRNA),CCK-8、划痕及Transwell实验检测细胞活力、迁移和侵袭能力.结果 GXYLT2在胃癌组织中比癌旁组织[(风险比(HR)=1.030,95%可信区间(CI)=0.690~1.370,P<0.05],并且其高表达与较差的总生存期(OS)(HR=2.450,95%CI:1.890~3.180)及无进展生存期(PFS)HR=2.780,95%CI:2.110~3.670)显著相关.此外,GXYLT2表达与多种肿瘤浸润免疫细胞,如B细胞[相关系数(r)=0.045]、CD4+T 细胞(r=0.352)、CD8+T 细胞(r=0.249)、中性粒细胞(r=0.323)、巨噬细胞(r=0.692)和髓样树突状细胞(r=0.360)等呈正相关.Cox回归分析表明,GXYLT2是胃癌的独立预后因素[CI:0.244(0.067~0.420)].体外实验证实,si-GXYLT2 组细胞迁移率[(60.44±3.52)%比(32.23±3.17)%,t=21.800,P<0.01]和侵袭数[(106.64±5.36)个 比(210.54±10.28)个,t=13.550,P<0.01]均低于Control GXYLT2组.结论 GXYLT2在胃癌中高表达与不良预后显著相关,并且可以影响胃癌细胞的迁移和侵袭能力.

Abstract

Objective To investigate the expression of glycosyltransferase 2(GXYLT2)in gastric cancer and its impact on patient prognosis,and to analyze its correlation with tumor-infiltrating immune cells and its effects on the biological characteristics of gastric cancer cells.Methods Based on the data in The Cancer Genome Atlas(TCGA)database,gastric cancer patients were classified into high/low expres-sion groups according to the median GXYLT2 expression,and independent samples t-test as well as chi-square test were used to analyze the expression of GXYLT2 in gastric cancer tissues and its relationship with patients'survival briefly supplemented with subgroup descriptions.The prognostic correlation between GXYLT2 and clinical characteristics was assessed by univariate and multivariate Cox proportional risk re-gression analysis.In vitro experiments were performed to divide HGC-27 gastric cancer cells into Control group(transfected with negative control siRNA)and si-GXYLT2 group(transfected with siRNA),and cell viability,migration,and invasive ability were detected by CCK-8,scratch,and Transwell assays.Results GXYLT2 was more prevalent in gastric cancer tissues than in paraneoplastic tissues[hazard ratio(HR)=1.030,95%confidence interval(CI):0.690-1.370,P<0.05]and its high expression was associated with poorer overall survival(OS)HR=2.450,95%CI:1.890-3.180)and progression-free survival(PFS)HR=2.780,95%CI:2.110-3.670)were significantly associated.In addition,GXYLT2 expression was positively correlated with a variety of tumor-infltrating immune cells,such as B cells[correlation coefficient(r)=0.045],CD4+T cells(r=0.352),CD8+T cells(r=0.249),neutrophils(r=0.323),macrophages(r=0.692)and myeloid dendritic cells(r=0.360).Cox regression analysis showed that GXYLT2 was an inde-pendent prognostic factor for gastric cancer[95%CI:0.244(0.067-0.420)].In vitro experiments confirmed that the cell migration rate[(60.44±3.52)%vs.(32.23±3.17)%,t=21.800,P<0.01]and the num-ber of invasion(106.64±5.36 vs.210.54±10.28,t=13.550,P<0.01)in the si-GXYLT2 group were lower than those in the control group.Conclusion The high expression of GXYLT2 in gastric cancer is sig-nificantly associated with poor prognosis and can influence the migration and invasion capabilities of gastric cancer cells.GXYLT2 may serve as a potential biomarker and therapeutic target for gastric cancer.
