Preliminary study on antibacterial,anti-inflammatory,and mosquito repellent activities of a compound traditional Chinese medicine essential oil
Objective To investigate the antibacterial and antiflammatory activities of a traditional Chinese medicine compound essential oil,evaluate its mosquito repellent efficacy,and analyze the chemical constituents and their relative contents.Methods The compound essential oil was extracted by steam distillation;the antibacterial activity of the compound essential oil against Staphylococcus aureus was determined by kirby-Bauer test;36 mice were randomly divided into a normal group,a model group,a Danpi phenol ointment group and a compound essential oil group,the mouse model of acute insect bite dermatitis was established,the pathological sections and inflammatory factor levels of mouse skin lesions were tested to verify the anti-inflammatory effect on insect bite dermatitis.The qualitative and quantitative analysis of the compound essential oil was conducted by gas chromatography-mass spectrometer(GC-MS).The mosquito repellent pharmacodynamics experiment of compound essential oil was carried out to verify its mosquito repellent effect.Results Compared with single essential oil,compound essential oil had a better inhibitory effect on the growth of Staphylococcus aureus,with a diameter of(17.25±0.37)mm as the antibacterial zone,which was moderately sensitive to Staphylococcus aureus;The compound essential oil effectively reduced the degree of skin tissue edema caused by insect bite dermatitis in mice,reduced the production of inflammatory cells and infiltration of mast cells in skin lesions,and lowered the serum TNF-α,IFN-γ,IL-6,IL-1β level in mice;The effective repellent time of compound essential oil against Aedes albopictus was 7.83 hours.A total of 28 chemical components such as aromatics and terpenoids were detected in the compound essential oil.Conclusion The compound essential oil has an inhibitory effect on the growth of gram positive bacteria and can effectively alleviate the skin inflammatory response caused by insect bite dermatitis in mice.Aromatic compounds account for the highest proportion of active ingredients,and the repellent time is superior,which meets the expected effect.
