Investigation on Japanese encephalitis virus carried by Culex tritaeniorhynchus in Sichuan Province
Objective To investigate the carrying situation of Japanese encephalitis virus of Culex tritaeniorhynchus in Sichuan Province, and to study the basic characteristics of the distribution of Japanese encephalitis virus,so as to provide a basis for predicting the epidemic risk of Japanese encephalitis and scientific prevention and control. Methods Samples of Culex tritaeniorhynchus were collected from rural areas of Sichuan Province from May to August 2023.RNA was extracted from a single female mosquito and reverse transcribed into cDNA.Japanese encephalitis virus in mosquito samples was detected by fluorescence quantitative RT-PCR based on TaqMan probe. The Chi-square test was used to compare the mosquito infection rate, and the Bonferroni test was used for pairwise comparison. Results A total of 1 070 mosquito samples were collected in 2023,of which 178 mosquito samples were positive for Japanese encephalitis virus, with an overall infection rate of 16.64%.According to the sampling area, the infection rate was higher in some cities in southern and eastern Sichuan.According to the sampling habitat, the infection rate was significantly different among different habitats(x2=27.012,P<0.05).The mosquito infection rate in pigsty(13.47%)was significantly lower than that in cattle pens(21.85%)and farmers(39.53%).According to the sampling month, the mosquito infection rate in May(30.56%)and August(26.21%)were significantly higher than that in June(13.03%)and July(6.68%).Conclusion The infection rate of Japanese encephalitis virus of Culex tritaeniorhynchus is high in Sichuan Province, and the rural area is an important natural focus of Japanese encephalitis virus.
