Intervention effect of astragalus polysaccharide on rats with Mycoplasma pneumoniae infection
OBJECTIVE To explore the intervention effect of astragalus polysaccharides on the rats with Mycoplas-ma pneumoniae infection and observe the impact on interleukin-6(IL-6)/phosphorylated signal transducer and ac-tivator of transcription 3(STAT3)signaling pathway.METHODS Totally 43 male SPF Wistar rats were enrolled in the study,10 of which were assigned as the blank group,and the rest of 33 were established for models of M.pneumoniae infection,30 rats were modeled successfully and were divided into the model group,the drug con-trol group and the astragalus polysaccharide group,with 10 rats in each group.The drug control group and the as-tragalus polysaccharide group were treated with montelukast sodium and astragalus polysaccharide,respectively;the blank group and the model group were treated with 0.9%sodium chloride solution.The pathological changes of the rats were observed by light microscope after the treatments for 7 days.The levels of inflammatory factors and oxidative stress indexes were detected by enzyme-linked immunosorbent assay(ELISA);the expression levels of IL-6 and STAT3 mRNA were detected by means of real-time fluorescent quantitative PCR;the relative expres-sion levels of IL-6/STAT3 signaling pathway-related proteins were detected by Western Blot(WB).RESULTS As compared with the blank group,the levels of tumor necrosis factor-α(TNF-α),C-reactive protein(CRP),ma-londialdehyde(MDA)and relative expression levels of IL-6 and STAT3 mRNA and proteins of the model group,the drug control group and the astragalus polysaccharide group were elevated,while the levels of interleukin-10(IL-10),glutathione peroxidase(GSH-Px)and superoxide dismutase(SOD)declined(P<0.05).The relative expression levels of TNF-α,CRP,MDA,IL-6,STAT3 mRNA and proteins of the drug control group and the as-tragalus polysaccharide group were lower than those of the model group,while the levels of IL-10,GSH-Px and SOD of the drug control group and the astragalus polysaccharide group were higher than those of the model group(P<0.05).The levels of TNF-α,CRP and MDA and relative expression levels of IL-6,STAT3 mRNA and pro-teins were higher in the astragalus polysaccharide group then in the drug control group,while the levels of IL-10,GSH-Px and SOD of the astragalus polysaccharide group were lower than those of the drug control group(P<0.05).CONCLUSION The astragalus polysaccharide may alleviate the inflammatory and oxidative stress response of the rats with M.pneumoniae infection and relieve the symptom of infection.The mechanism may be associated with the inhibition of IL-6/STAT3 signaling pathways.
