目的 对肺炎支原体感染大鼠进行鱼腥草提取物治疗,对其所产生抗炎功效及作用机制进行分析。方法 SPF级Wistar雄性大鼠分为空白组、肺炎支原体感染模型组、鱼腥草提取物低剂量组和高剂量组各9只。空白组、模型组只进行生理盐水灌胃,低剂量组和高剂量组给予鱼腥草提取物125 mg/kg、500 mg/kg灌胃,持续7 d后检测外周血转化生长因子-β(TGF-β)、涎液化糖链抗原-6(KL-6)、表面活性蛋白D(SP-D)、炎症因子及Toll样受体4(TLR4)、髓样分化因子88(MyD88)、核因子κB(NF-κB)mRNA相对表达量,免疫印迹法检测肺组织中TLR4、MyD88、NF-KB蛋白表达。结果 与空白组比较,模型组、鱼腥草提取物低剂量组、高剂量组TGF-β、KL-6、SP-D、IL-13、TNF-α 水平、TLR4、MyD88、NF-κB mRNA 表达量、TLR4、MyD88、NF-κB 蛋白表达量均升高,IL-12 水平降低(P<0。05);与模型组、低剂量组比较,高剂量组TGF-β、KL-6、SP-D、IL-13、TNF-α水平、TLR4、MyD88、NF-κB mRNA表达量、TLR4、MyD88、NF-KB蛋白表达量均降低,IL-12水平升高(P<0。05)。结论 通过鱼腥草提取物对肺炎支原体感染大鼠进行治疗干预,以改善局部乃至支气管和肺组织炎症反应,达到较好的干预效果。
Anti-inflammatory effect of Houttuynia extract on rats with Mycoplasma pneumoniae infection and mechanisms of action
OBJECTIVE To observe the anti-inflammatory effect of Houttuynia extract on rats with Mycoplasma pneumoniae infection and analyze the mechanisms of action.METHODS The Wistar rats of SPF grade were divid-ed into the blank group,the M.pneumoniae infection model group,the low-dose houttuynia extract group and the high-dose houttuynia extract group,with 9 rats in each group.The blank group and the model group were only treated with normal saline for gavage,the low-dose group and the high-dose group were respectively treated with 125 mg/kg and 500 mg/kg of houttuynia extract for gavage.The levels of peripheral blood transforming growth factor-β(TGF-β),Krebs Von den Lungen-6(KL-6),surface active protein D(SP-D),inflammatory factors and the relative expression levels of Toll-like receptor 4(TLR4),myeloid differentiation factor 88(MyD88)and nu-clear factor κB(NF-κB)mRNA were detected after the treatments for 7 consecutive days;the expression levels of TLR4,MyD88 and NF-κB proteins in lung tissues were detected by Western Blot.RESULTS The levels of TGF-β,KL-6,SP-D,IL-13,TNF-α,the expression levels of TLR4,MyD88,NF-κB mRNA,and the expression lev-els of TLR4,MyD88 and NF-κB proteins were higher in the model group,the low-dose houttuynia extract group and the high-dose houttuynia extract group than in the blank group,while the IL-12 level was lower in the model group,the low-dose houttuynia extract group and the high-dose houttuynia extract group than in the blank group(P<0.05).The levels of TGF-β,KL-6,SP-D,IL-13,TNF-α,the expression levels of TLR4,MyD88,NF-κB mRNA,and the expression levels of TLR4,MyD88 and NF-κB proteins were lower in the high-dose houttuynia extract group than in the model group and the low-dose houttuynia extract group;the IL-12 level of the high-dose houttuynia extract group was higher than that of the model group and the low-dose houttuynia extract group(P<0.05).CONCLUSION As a treatment intervention,Houttuynia extract may alleviate the local inflammatory response or even the inflammatory responses in bronchia and lung tissues so as to achieve favorable intervention effect.
