目的 探讨胃癌组织中壳多糖酶3样蛋白1(CHI3L1)、核富集转录体1(NEAT1)表达及其与乙型肝炎病毒(HBV)感染的关系。方法 回顾性选取202例于2018年6月-2023年3月在重庆市綦江区人民医院内镜中心进行检查的胃癌患者的临床资料,收集胃癌组织及癌旁组织,分为胃癌组(n=202)和癌旁组(n=202),根据是否HBV感染分为HBV(+)和HBV(-);比较胃癌组和癌旁组中CHI3L1、NEAT1表达及HBV感染情况,CHI3L1、NEAT1表达与胃癌患者临床病理参数的关系;分析患者胃癌组织CHI3L1、NEAT1表达与HBV感染的相关性。结果 胃癌组CHI3L1、NEAT1阳性表达率高于癌旁组(P<0。05);在胃癌组中,HBV阳性患者CHI3L1、NEAT1阳性表达率高于HBV阴性患者(P<0。05);胃癌组不同TNM分期患者中,Ⅲ~Ⅳ期患者的NEAT1阳性表达率较Ⅰ~Ⅱ期患者高(P<0。05);有淋巴结转移的CHI3L1、NEAT1阳性表达率较无淋巴结转移高(P<0。05);分化程度中,低分化的CHI3L1阳性表达率较中高分化低(P<0。05);肿瘤浸润深度中,侵犯浆膜及浆外层的CHI3L1、NEAT1阳性表达率较侵犯黏膜及肌层高(P<0。05);胃癌组CHI3L1、NEAT1表达与HBV感染呈正相关(r=0。431;0。479,P<0。05)。结论 胃癌患者癌组织中CHI3L1、NEAT1呈高表达,CHI3L1表达可能与淋巴结转移、分化程度、肿瘤浸润深度有关,NEAT1表达可能与肿瘤TNM分期、淋巴结转移、肿瘤浸润深度有关;胃癌组织中CHI3L1、NEAT1阳性表达情况与HBV感染呈正相关。
Expressions of CHI3L1 and NEAT1 in gastric cancer tissues and association with HBV infection
OBJECTIVE To investigate the expression of chitosanase 3-like protein 1(CHI3L1)and nuclear enriched transcript 1(NEAT1)in gastric cancer tissues and their relationship with hepatitis B virus(HBV)infection.METHODS Clinical data of 202 patients with gastric cancer who underwent examined at the Endoscopy Center of Qijiang District People's Hospital of Chongqing from Jun.2018 to Mar.2023 were retrospectively selected,and the gastric cancer tissues and adjacent tissues were collected and divided into gastric cancer group(n=202)and adjacent cancer group(n=202),which were divided into HBV(+)and HBV(-)according to whether they were infected with HBV.The expression of CHI3L1,NEAT1 and HBV infection in gastric cancer group and para-cancer group,as well as and the relationship between the expression of CHI3L1 and NEAT1 and clinicopathologi-cal parameters of gastric cancer patients was compared.The correlation between the expression of CHI3L1 and NEAT1 in gastric cancer tissues and HBV infection was analyzed.RESULTS The positive expression rates of CHI3L1 and NEAT1 in the gastric cancer group were higher than those in the adjacent cancer group(P<0.05).In gastric cancer group,the positive expression rate of CHI3L1 and NEAT1 in HBV-positive patients was higher than that in HBV-negative patients(P<0.05).Among patients with different TNM stages in the gastric cancer group,the positive expression rate of NEAT1 was higher in patients with stage Ⅲ to Ⅳ than in patients with stage Ⅰ to Ⅱ(P<0.05).The positive expression rates of CHI3L1 and NEAT1 in patients with lymph node me-tastasis were higher than those without lymph node metastasis(P<0.05).In the degree of differentiation,the positive expression rate of CHI3L1 in low-differentiation was significantly lower than that in middle and high dif-ferentiation(P<0.05).In the depth of tumor invasion,the positive expression rates of CHI3L1 and NEAT1 in the invasion of the plasma membrane and outer plasma layer were higher than those in the invasion of the mucosa and muscle(P<0.05).The expression of CHI3L1 and NEAT1 in gastric cancer group was positively correlated with HBV infection(r=0.431,0.479,P<0.05).CONCLUSION The expression of CHI3L1 and NEAT1 was highly expressed in cancer tissues of gastric cancer patients,and the expression of CHI3L1 might be related to lymph node metastasis,differentiation degree,and depth of tumor invasion,while the expression of NEAT1 might be related to tumor TNM stage,lymph node metastasis,and depth of tumor invasion.The positive expres-sion of CHI3L1 and NEAT1 in the tissues of gastric cancer was positively correlated with HBV infection.
