丹参提取物对感染性骨缺损大鼠炎症水平的影响

目的 分析丹参提取物对感染性骨缺损大鼠炎症水平的影响。方法 选取50只SPF级SD大鼠,10只大鼠作为对照组,另外40只建立感染性骨缺损模型,将30只建模成功大鼠分为模型组、药物对照组、丹参提取物组各10只;观察各组大鼠病理组织学检测、骨密度、骨灰度、血管内皮生长因子(VEGF)、骨生长因子、炎症因子水平、降钙素基因相关肽(CGRP)/磷脂酰肌醇-3激酶(PI3K)/丝氨酸苏氨酸蛋白激酶(Akt)通路相关mRNA表达量、CGRP/PI3K/Akt通路。结果 与对照组相比,模型组、药物对照组、丹参提取物组骨密度、骨灰度、VEGF、胰岛素样生长因子-1(IGF-1)、碱性成纤维细胞生长因子(FGF-2)、CGRP、PI3K、Akt mRNA表达量、CGRP、PI3K、Akt表达量降低,白细胞介素-10(IL-10)、肿瘤坏死因子-a(TNF-a)、核因子κB(NF-κB)升高(P<0。05);与模型组相比,药物对照组、丹参提取物组骨密度、骨灰度、VEGF、IGF-1、FGF-2、CGRP、PI3K、Akt mRNA表达量、CGRP、PI3K、Akt表达量升高,IL-10、TNF-a、NF-KB降低(P<0。05);与药物对照组相比,丹参提取物组骨密度、骨灰度、VEGF、IGF-1、FGF-2、CGRP、PI3K、Akt mRNA 表达量、CGRP、PI3K、Akt 表达量升高,IL-10、TNF-a、NF-κB 降低(P<0。05)。结论 采用丹参提取物对感染性骨缺损大鼠干预,可使大鼠体内血管内皮生长因子、骨生长因子表达升高,并减轻炎症因子水平,促使大鼠的恢复,其作用机制可能与调控CGRP/PI3K/Akt通路有关。
Effect of salvia miltiorrhiza extracts on level of inflammation in rats with infected bone defects
OBJECTIVE To investigate the effect of salvia miltiorrhiza extracts on the level of inflammation in rats with infected bone defects.METHODS Fifty SPF-grade SD rats were selected,10 rats were used as the control group,and another 40 rats were modeled as infected bone defects.Thirty rats with successful modeling were divid-ed into the model group,the drug control group,and the radix salviae miltiorrhizae extract group,with 10 rats in each group.The pathological histology,bone mineral density,bone ash content,levels of vascular endothelial growth factor(VEGF),bone growth factor,inflammatory factors,expression of calcitonin gene-related peptide(CGRP)/phosphatidylinositol 3-kinase(PI3K)/serine threonine protein kinase(Akt)pathway-related mRNA ex-pression and CGRP)/PI3K/Akt pathway in the rats of all the groups were observed.RESULTS Compared with the control group,bone mineral density,bone ash content,vascular endothelial growth factor(VEGF),insulin-like growth factor-1(IGF-1),basic fibroblast growth factor(FGF-2),CGRP,PI3K,Akt mRNA expression,CGRP,PI3K,Akt expression were decreased in the model group,the drug control group,and the Radix Salviae Miltior-rhizae extract group,while the interleukin-10(IL-10),tumor necrosis factor-a(TNF-a),and nuclear factor κB(NF-κB)were increased(P<0.05).Compared with the model group,the bone mineral density,bone ash con-tent,VEGF,IGF-1,FGF-2,CGRP,PI3K,Akt mRNA expression,CGRP,PI3K,Akt expression levels in the medication control group and the Radix Salviae Miltiorrhizae extract group were increased,while IL-10,TNF-αand NF-κB were decreased(P<0.05).Compared with the drug control group,the bone mineral density,bone ash content,VEGF,IGF-1,FGF-2,CGRP,PI3K,Akt mRNA expression,CGRP,PI3K,Akt expression were in-creased in the Radix Salviae Miltiorrhizae extract group,while IL-10,TNF-α and NF-κB were decreased(P<0.05).CONCLUSION The intervention of salvia miltiorrhiza extract on rats with infected bone defects elevated the expression of vascular endothelial growth factor and bone growth factor in rats,attenuated the level of inflamma-tory factors,and promoted the recovery of rats,and its mechanism might be related to the regulation of CGRP/PI3K/Akt pathway.
