黄牛奶树树茎醇提取物内服对狼疮肾炎并发尿路感染患者JAKSTAT信号通路的影响

目的 分析黄牛奶树树茎醇提取物对Janus激酶信号转导子与转录激活子途径(JAKSTAT)信号通路狼疮肾炎并发尿路感染的影响。方法 选取2022年6月—2023年6月于四川大学华西医院接受诊治的狼疮肾炎并尿路感染患者45例纳为研究对象,分成模型组、低剂量组、高剂量组各15例,另选同期于医院体检的健康者30名作为对照组;对照组和模型组不做处理,低剂量组、高剂量组分别给予15 g、30 g黄牛奶树树茎醇提取物调控;观察患者系统性红斑狼疮活动指数(SLEDAI)评分、24 h尿蛋白、不同因素尿路感染情况、尿路感染患者尿培养结果、血肌酐(Scr)、补体3(C3)、C4、血沉(ESR)、降钙素原(PCT)、超敏C-反应蛋白(hs-CRP)、白细胞(WBC)、白细胞介素-12(IL-12)、转化生长因子β1(TGF-β1)、IL-6水平。结果 45例尿路感染患者检出病原菌以大肠埃希菌为主;与对照组相比,模型组、低剂量组、高剂量组Scr、ESR、PCT、hs-CRP、WBC、IL-12、TGF-β1、IL-6均升高,C3、C4 下降(P<0。05);与模型组相比,低剂量组、高剂量组 Scr、ESR、PCT、hs-CRP、WBC、IL-12、TGF-β1、IL-6呈下降,而C3、C4呈升高(P<0。05);与低剂量组相比,高剂量组Scr、ESR、PCT、hs-CRP、WBC、IL-12、TGF-β1、IL-6水平均降低,C3、C4升高(P<0。05)。结论 黄牛奶树树茎醇提取物是通过调节JAKSTAT信号通路,抑制炎症因子,平衡免疫功能,提高预防尿路感染的作用。
Effect of oral administration of alcohol extract of yellow milk tree stems on JAKSTAT signaling pathway of lupus nephritis patients complicated with urinary tract infection
OBJECTIVE To analyze the effects of alcohol extract of yellow milk tree stems on Janus kinase signal transductor and activator of transcription pathway(JAKSTAT)signaling pathway in patients with lupus nephritis complicated with urinary tract infection.METHODS A total of 45 patients with lupus nephritis and urinary tract infection who were diagnosed and treated at the West China Hospital of Sichuan University from Jun.2022 to Jun.2023 were selected as the study subjects,and were divided into the model group,the low-dose group and the high-dose group,with 15 cases in each group,and 30 healthy patients who underwent physical examination in the hospital during the same period were selected as the control group.The control and model groups were not trea-ted,and the low-dose and high-dose groups were given 15 g and 30 g of alcohol extract of of yellow milk tree stems,respectively.The SLE activity index(SLEDAI)score,24h urinary protein,urinary tract infection with different factors,urinary culture results,serum creatinine(Scr),complement C3(C3),C4,erythrocyte deposi-tion rate(ESR),procalcitonin(PCT),hypersensitive C-reactive protein(hs-CRP),and white blood cells(WBC),interleukin-12(IL-12),transforming growth factor β1(TGF-β1),interleukin-6(IL-6)levels were ob-served.RESULTS Escherichia coli was the main pathogen detected in 45 cases of urinary tract infection.Com-pared with the control group,Scr,ESR,PCT,hs-CRP,WBC,IL-12,TGF-β1 and IL-6 were elevated in the model,low-dose and high-dose groups,while C3 and C4 were decreased(P<0.05).Compared with the model group,Scr,ESR,PCT,hs-CRP,WBC,IL-12,TGF-β1 and IL-6 in low-dose and high-dose groups were de-creased,while C3 and C4 were increased(P<0.05).Compared with the low-dose group,the levels of Scr,ESR,PCT,hs-CRP,WBC,IL-12,TGF-β1 and IL-6 in high-dose group were decreased,while C3 and C4 were in-creased(P<0.05).CONCLUSION The alcoholic extract of yellow milk tree stem was known to inhibit inflamma-tory factors,balance immune function,as well as improve the prevention of urinary tract infection by modulating theJAKSTAT signaling pathway.
